非熱大氣電漿處理傷口癒合之評估

摘要


非熱大氣電漿產生的活性氧物種(reactive oxygen species, ROS),在醫療的應用上成為一種有效的工具,包含血液凝固、滅菌和惡性腫瘤凋亡。電漿已被證明可以利用ROS誘導纖維母細胞因子-2釋放使內皮細胞促進血管新生。低劑量的ROS透過生長因子相關機制,可以增加內皮細胞的遷移、增生及血管形成。反之,高劑量的ROS會造成細胞傷害甚至凋亡。因此本研究目的利用自行開發設計的非熱噴射式大氣電漿系統應用於傷口處理,調控傷口累積的ROS劑量來改變傷口癒合速率。電漿設備在29.4 kV的工作電壓與116.6 kHz的工作頻率下激發電漿,此激發狀態的電流為0.13 A,消耗功率為20.57 W,氣體流量5 slpm,電漿激發最大長度為30 mm,激發過程中最高溫度為30 度。本研究中使用大腸桿菌與金黃色葡萄球菌做為滅菌效能的檢測,於5分鐘電漿治療可降低細菌量達到50 %以上。實驗結果顯示本電漿系統確實可以產生ROS,例如單態氧、氫氧自由基,並對組織殺菌與傷口癒合有顯著影響。非熱電漿可加速傷口癒合的不同時期,抑制細菌定植,並最終達到促進傷口癒合。

Non-thermal atmospheric pressure plasma produces reactive oxygen species (ROS), which has recently become an efficient tool in medical applications including blood coagulation, bacterial sterilization and malignant cell apoptosis. The plasma is demonstrated to promote angiogenesis through the release of fibroblast growth factor-2 induced by ROS. Low ROS doses may increase endothelial cell migration, proliferation and tube formation through growth factor-related mechanisms. Conversely, high ROS doses can cause cell damage or apoptosis. The objective of this study is to alter the wound treating rate of full-thickness skin on Wistar rats by adjusting the ROS doses with a self-made non-thermal atmospheric pressure plasma jet generator. The applied voltage and the discharge current of a damping waveform is respective 29.4 kV and 0.13 A, the frequency is 116.6 kHz, the total power of plasma-jet is 20.57 W, the maximum plasma length and applied temperature is respective 30 mm and 30 oC at gas flow 5 slpm. The quantitative analysis result of E. coli and S. aureus sterilization that the 5 min of plasma treatment reduced the number of CFU/ml at least 50%. This result can be related to the presence in the plasma volume of active species, such as O, OH radicals. This study will help identify the impact of plasma exerting on tissue sterilization and wound healing. Non-thermal plasma can accelerate the different phase of wound healing, suppress the bacteria colonization and eventually promote the wound healing.
