蓮霧葉部精油之碳氫化合物成分分析

摘要


蓮霧(Syzygium samarangense (Blume) Merr .et Perry)為桃金孃科赤楠屬的常綠小喬木植物,將新鮮採集的蓮霧葉部,經清洗、風乾與攪碎等步驟後進行水蒸汽蒸餾以帶出揮發性精油成分,經溶劑乙醚和正己烷萃取、鹼處理,去水減壓濃縮後,得一淡黃色之中性精油。 將中性精油先以管柱色層分析法粗分成碳氫化合物與含氧化合物兩大部分,再分別以氣相色層分析-火焰離子偵測器(GC-FID)及氣相色層分析-質譜儀(GC-MS)進行光譜鑑定,初步瞭解精油大致的成分;另再以管柱色層分析進行單離與純化,然後測其傅立葉轉換紅外光光譜儀(FT-IR)、核磁共振波譜儀(NMR)進行化學結構分析,或予以之標準物質GC-Rt值及文獻光譜核對,以鑑定中性精油其所含化學成分,其中由碳氫化合物中單離出三個化合物α-pinene、o-cymene、caryophyllene,而以GC-MS光譜鑑定15個化合物α-thujene、camphene、β-pinene、limonene、γ-terpinene、δ-terpinene、ylangene、copaene、β-elemene、6,6,10,10-tetramethyl-2- methylenebicyclo[7.2.0]undecane、β-selinene、α-selinene、τ-muurolene、calamenene、cadina-1,4-diene 。

The wax apple (Syzygium samarangenes (Blume) Merr. et Perry) was an evergreen small tree which belonged to genus Syzygium of the Myrtaceae family. This study collected the fresh leaves of the wax apple and obtained the volatile components of essential oil through the process of cleansing, drying, mincing and steaming. From the components, this study furthered to obtain the light yellow neutral essential oil through the extraction by ether solvent and n-Hexane, alkali treatment, dehydration and vacuum concentration. First of all, the neutral oil was roughly separated into hydrocarbon and oxygenates by column chromatography. Secondly, the main components of the essential oil were analyzed respectively by gas chromatography-GC-FID and GC-MS. This study furthered to isolate and purify the components by column chromatography, and then estimated and analyzed the chemical structures by fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (NMR), or verified the chemical structures with the GC-Rt values of standard substance and the spectrum in the literature, in order to identify the chemical components of the neutral essential oil. In the result of this study, the components were isolated as α-pinene, o-cymene, caryophyllene and were identified by GC-MS as 15 chemical compounds – α-thujene、 camphene、 β-pinene、 limonene、 γ-terpinene、 δ-terpinene、 ylangene、copaene、β-elemene、6,6,10,10-tetramethyl-2-methylenebicyclo[7.2.0]undecane、 β-selinene、α-selinene、τ-muurolene、calamenene、cadina-1,4-diene.
