摘要 本論文研究是為了求得血管中血球的流動速度,使用了攜帶較方便的光學顯微鏡,加上CCD攝影機連接影像擷取卡,將動態影像儲存至電腦硬碟中;觀察並且取得影像的活體目標是後腿背上植入癌細胞的裸鼠。 因為是活體顯微實驗,因此在麻醉過後,裸鼠的呼吸或抖動都會對影像造成頗大的影響,所以在計算血流速度之前,我們將原始影像做了對位處理。解決了影像對位問題後,將想要取得血流速度的血管中心線部分,按照時間的順序,可得到血管中心線的血流profile,再利用Hough Transform 計算出血流profile圖上的斜率,而這些斜率的倒數即是血流速度。 因此,透過本論文的實驗分析即可得到裸鼠腫瘤上表層的血管分佈、血流速度的變化。將有助於裸鼠相關實驗血流速度的觀察與分析。
Abstract This study is to derive from the speed that blood cells flow in blood vessel. This study uses optic microscope which is convenient to take along plus CCD video camera which connect with video capture card store up develop images into computer hard discs. The live target for observe and get mages is nude mice which embedding cancer cell in back of hind leg. This study is an Intravital microscopy so that after anesthesia the breath or shake from nude mice will effect the image. For this reason before counting the speed of blood stream we deal with original image by using image registration. After solve the problem of image registration, the center line of blood vessel which we want to acquire the speed of blood stream will according to the order of time get the blood stream profile. Then, using Hough Transform counting the slope of picture of blood stream the speed of blood stream will be the reciprocal of slope. Therefore, by the experimental analysis of this study could understand distribute of blood vessel, the change of speed of blood stream on the surface of tumor of nude mice. This study will help to observe and analysis those experiments which are related to nude mice of speed of their speed of blood stream.
