脫羧基凝血酶原生物感測器應用於早期肝癌檢測之研發

摘要


根據台灣行政院衛生署所公布的統計資料顯示,台灣國人每年大約有七千名患者因肝癌而死亡,肝癌在台灣癌症死亡率排名第二,肝癌對國人的健康已經造成重大的影響。 本研究目的為研發一種能即時量測肝癌標的物-脫羧基凝血酶原DCP之阻抗變化生物感測器,使用網版印刷技術,將銀膠線路與保護銀膠線路的碳膠層印製於感測器基材上,外層則由絕緣層覆蓋並繪製感測窗口,最後再將脫羧基凝血酶原DCP(Des-gamma-carboxy prothrombin)抗體固定於感測器上製作出DCP生物感測器。 經由阻抗分析儀量測過後經由數據處理後發現DCP阻抗分析於16.4kHz~73.1kH擁有良好的線性度R2>0.99,且在此頻寬中於 27kHz呈現最佳線性度R2=0.9953,另外加入電容值與電導值量測,經數據處理分析後發現電容值於4.47kHz~8.83kHz擁有良好的線性度R2>0.98,且在此頻寬中於6.08kHz有最佳線性度R2=0.9961,而電導值也於2.55 kHz -7.74 kHz頻段也有線性度R2>0.9,且此頻段中於2.89kHz也出現最佳線性度R2=0.9917。 在未來可配合讀出電路使其可達到方便隨身攜帶,快速且可以及時得知量測之結果,並達到居家照護、預防與即時監測肝癌與早期發現與早期治療之目的。

According to the C.D.C statistics in 2013, liver cancer held the second position of the top leading causes of cancer death and this caused a huge damage to human health. The purpose of this study was to design a Des-gamma-carboxy prothrombin immunobiosensor for liver cancer screening in its early stage. Screen-printing technique was employed in this study, with the PET plastic as the immunobiosensor substrate. Silver paste was printed as the conduction lines and graphite paste was printed as the electrode pads. The DCP antibody was fixed onto the sensor. Result showed that the biosensor performs good linearity in the frequency range of 16.4KHz ~ 73.1KHz, with the best linearity R2=0.9953 at 27kHz. By analyzing the capacitance and conductive response, results showed that the capacitance response performs a good linearity in the frequency range of 4.47kHz ~ 8.83kHz, with the best linearity R2=0.9961 at 6.08kHz, and the conductive response performs a good linearity in the frequency range of 2.55kHz ~ 7.74kHz, with the best linearity R2=0.9917 at 2.89kHz. If a readout circuit is connected with the immunobiosensor, the immunobiosensor can then be used at home to provide a rapid and simple screening of liver cancer in its early stage.
