摘要


隨著生活型態的改變,缺乏規律的運動、不健康的飲食習慣以及極端的氣候型態,都是誘發心血管疾病(Cardiovascular diseases,CVD)的危險因素。雖然心血管疾病較常見於老年人或特定族群,但若缺乏平日保健預防,也可能會因急性發作造成心肌梗塞及腦中風,因此定期健康檢查相當重要。 高水平的高密度脂蛋白膽固醇(High-density lipoprotein cholesterol,HDL-C)一直被認為具有預防心血管的作用,但目前研究顯示HDL-C並不是一個好的量測心血管疾病風險的指標。載脂蛋白E(Apoliporotein E,ApoE)在脂質代謝中扮演重要角色,是作為低密度脂蛋白受體(Low-density lipoprotein receptor,LDLR)的配體,能維持體內脂質的平衡。而含有ApoE的HDL-C與總HDL-C比值與動脈粥樣硬化(Atherosclerosis)有較大的相關性,因此可以ApoE-HDL-C/HDL-C作為心血管疾病的指標。 本研究研發適體電化學感測器結合酵素反應,將ApoE適體(Aptamer)修飾於拋棄式網版印刷碳電極(Screen-printed carbon electrode,SPCE),透過適體專一性辨識含有ApoE的HDL,並進行電極與實驗條件的最佳化,量測人體中含有ApoE的HDL-C與總HDL-C的比值(ApoE-HDL-C/HDL-C)。本設計預期可應用於評估心血管疾病的風險。

With the lifestyle change, the lack of regular exercises, unhealthy eating habitat and the extreme weather are the risk factors that cause the induction of cardiovascular diseases (CVD). Not only elders and disease-specific group especially vulnerable to CVD, neglect of maintaining in a healthy state may also lead to CVD or stroke due to acute attack. Therefore, it is essential to have a periodic health examination. High levels of high-density lipoprotein cholesterol (HDL-C) have been considered to prevent CVD, but some studies show that HDL-C is not a reliable indicator of the risk of cardiovascular disease. Apolipoprotein E (ApoE) plays an important role in lipid metabolism and acts as a ligand for low-density lipoprotein receptor (LDLR) to maintain lipid homeostasis. As ApoE-HDL-C/HDL-C ratio is associated with atherosclerosis, it can be used as an indicator of CVD. We developed a DNA aptamer electrochemical sensor to specifically recognize ApoE-HDL. Aptamer modified screen printed carbon electrode (SPCE) were loaded with HDL. ApoE-included HDL were remained after washes. The cholesterol content was then evaluated by measuring hydrogen peroxide content generated by the reaction of enzyme. The concentration of surfactant, cholesterol estersase and cholesterol oxidase were optimized. Based on this design, the determined ratio of ApoE-HDL-C to HDL-C were potentially applicable for the evaluation of the risk of CVD.
