摘要


本論文利用生物感測器之即時性(real-time)、高靈敏性(high sensitivity)、高選擇性(high selectivity)等特性,結合奈米粒子特殊的生物相容性與物理特性,使用於生物辨識偵測及製作感測平台。實驗使用網版印刷方式,印製銀膠(Sliver Plastic),碳膠(Carbon Plastic)及絕緣膠(Dielectric Plastic)在基板上製作生物感測器與其電極,為了提升感測器靈敏度加入奈米粒子氧化鋅(Zinc Oxide)及氧化鋁(Aluminum Oxide)。生物辨識載體選擇新型肝癌標的物,脫羧基凝血酶(De-Gamma-Carboxy Prothrombin,DCP)抗原及抗體,藉由生物感測器電極量出之阻抗值,經計算扣除阻抗效應後得到相應電容的改變來檢測DCP濃度的變化,並在不同頻率下測量阻抗的變化以選出最佳頻率,實驗主要針對不同奈米顆粒對於自製生物感測器特性的影響,與其未來應用於生物醫學偵測與診斷。

A Des-Gamma-Carboxy Prorhrombin(DCP) biosensor is a device that detects changes in cell impedance, and which is employed for screening of early-stage liver cancer medical treatment. A biosensor is a device designed to detect or quantify a biochemical molecule such as a particular protein or DNA sequence. Impedance biosensors for simple, rapid, label-free, low-cost detection of biomolecules. Normalization of Des-gamma-carboxy prothrombin(DCP) levels correlates well with successful tumor resection and appears to be an excellent marker of tumor activity. DCP has about 70% sensitivity and almost 100% specificity for liver cancer detection. DCP screening cannot be done at home because sophisticated instruments are needed for the detection of very low concentration DCP. The blood was measured by impedance biosensor, which can obtain impedance value and then determine whether it suffers cancer or not. This study is to develop a home-used and point-of-care immunobiosensor for the screening of early-stage liver cancer in order to achieve the goal of early medical treatment.
