以液相層析電噴灑四極柱飛行時間質譜儀及毛細管電泳線上濃縮技術對九種色胺類濫用藥物的分析研究

摘要


「狡詐家藥物」是指用來規避現有法律規範所合成出的一系列化合物。本實驗所分析的化合物為九種色胺類濫用藥物,由於九種分析物的化學結構非常相似且能夠允許在生物檢體中的含量非常微量。因此,為了有效的對九種色胺類濫用藥物進行分析,應採用同時具有高準確度及高靈敏度的分析方法。本研究中,使用液相層析電噴灑飛行時間質譜儀 (liquid chromatography/electrospray ionization quadrupole time-of-flight mass spectrometry, LC-Q-TOFMS)及毛細管電泳線上濃縮技術對有九種色胺類濫用藥物進行分離及偵測。真實樣品部分,為無不良嗜好健康捐贈者的唾液檢體,將九種色胺類濫用藥物額外添加至唾液檢體中,經由液液萃取對唾液檢體進行前處理,再以最佳化液相層析電噴灑飛行時間質譜儀條件對唾液檢體進行檢測。最後,我們使用了市售的MassWorks軟體來改善質譜的精確度和分辨率,也將同位素校正原理應用在LabVIEW (Laboratory Virtual Instrumentation Engineering Workbench)程式設計軟體上,並比較兩者的校正結果。

Designer drug is a term of synthesized compounds that are used to get around existing drug laws. For the analysis of these compounds, the method used should have a high degree of accuracy and a high sensitivity, not only because their chemical structures are similar, but also because their levels in biological fluids are usually very low. In this study, optimal conditions for the separation and detection of a mixture of 9 tryptamine designer drugs are described, by the use of liquid chromatography/electrospray ionization quadrupole time-of-flight mass spectrometry (LC-Q-TOFMS) and capillary electrophoresis/UV absorbance in conjunction with online sample concentration techniques. Saliva samples were obtained by spiking saliva from volunteers with the 9 tryptamine designer drugs. After liquid-liquid extraction, the samples were examined by LC-Q-TOFMS method. The extraction procedures used for the saliva sample and the experimental conditions are reported in detail. Furthermore, a commercial program, MassWorks software, was applied to improve the mass accuracy and resolution. With respect to precision and simplicity of use, a homebuilt mass calibration program by means of LabVIEW (Laboratory Virtual Instrumentation Engineering Workbench) was also used to compare to the results obtained from the MassWorks.
