白千層成分分析及抗氧化活性之探討

摘要


白千層(Melaleuca leucadendra)屬於桃金孃科(Myrtaceae),白千層屬(Melaleuca)的植物,主要分佈於澳大利亞、印尼等地。白千層為防風林的主要樹種,具有耐鹽、耐旱等特性,故在沿海一帶常種植。本研究是探討白千層精油成分分析和抗氧化活性,開發白千層的附加價值,為沿海農民帶來新的商機。 樺木酸(betulinic acid)是一種萃取自白樺樹的五環三萜類化合物 (pentacyclic triterpenoid),具有抗腫瘤、抗病毒和抗發炎的活性,其中以抗癌活性而聞名,經文獻發現,白千層葉子含有豐富的樺木酸,具有抗炎活性成分,對未來保健醫療有很大的效益。 本實驗採用一般蒸氣蒸餾法(SD)、減壓蒸氣蒸餾法(VSD)及超音波萃取法(UE)來提煉白千層葉部,再利用氣相層析儀(GC)來檢測精油成分及利用高液相色譜法(HPLC)檢測樺木酸含量。再檢測其清除DPPH清除自由基能力、總多酚含量、及還原力。 結果顯示,經GC檢測後,雲林元長鄉所種植的白千層主要以1,8-Cineole為主;其次為Limonene。經HPLC檢測後,白千層含有豐富的樺木酸,代表著白千層也具有抗炎抗癌的效果。一般蒸餾法蒸餾出的白千層精油以編號5擁有最好的抗氧化能力,以減壓蒸餾法蒸餾出的白千層精油以編號2擁有最好的抗氧化能力,以超音波萃取法萃取出來的白千層粗萃物以編號2擁有最好的抗氧化能力,最佳化以條件75%/40m/180W/60℃含有最多的樺木酸含量,抗氧化能力也較佳,其發現清除DPPH能力越佳總多酚含量也越多,還原能力也比較好,還原力越強,同時也表示供給電子能力越佳。 綜合以上結果,白千層含豐富的1,8-Cineole以及抗氧化能力,若能取代人工抗氧化劑,將可用於日常生活中,提高白千層附加價值。

Melaleuca leucadendra belongs to Myrtaceae, Melaleuca is mainly distributed in Australia, Indonesia and Asia. Melaleuca trees is a typical windbreak plants, it can tolerate high salt, concentration and drought; therefore, it is often planted in the coastal areas. The main objectives of this investigation was to extract essential oils from Melaleuca leucadendra by steam distillation (SD), vacuum steam distillation (VSD) and ultrasonic-assisted extraction (UAE) methods, adding value and daily commodities. Betulinic acid is a pentacyclic triterpenoids confession birch extract, have anti-tumor, anti-viral and anti-inflammatory activities,the Other researches found that white thousand layer birch leaves contain rich acid, and has anti-inflammatory active ingredient This characteristic could be beneficial to the future of health care. In this study,essential oil components were identified by the gas chromatography (GC) and High Pressure Liquid Chromatography (HPLC) ; antioxidant activities were performed with total phenolic content assays, DPPH free radical scavenging assay and reducing power assay. GC analysis of the essential oils showed that the majority components of the essential oil were 1,8-cineole . HPLC analysis of the essential oils also showed that the Melaleuca has rich betulinic acid. Steam distillation (SD) has best Antioxidant capacity in YC-5, and vacuum steam distillation (VSD) has best Antioxidant capacity in YC-2, Ultrasonic-assisted extraction (UAE) has best Antioxidant capacity in YC-2,In75%/40m/180W/60℃condition and is richer in betulinic acid than other condition
