飼料中添加青海菜對日本鰻肝臟中抗氧化酵素之影響

Effects of Monostroma nitidum powder supplement on the antioxidant enzymes in liver of Japanese eel (Anguilla japonicus)

摘要


本研究目的主要為探討青海菜在日本鰻飼料中不同比率添加量(分別添加0、3.0、6.0 和9.0%藻粉)對其成長表現及肝臟中抗氧化與解毒酵素活性之影響,與是否降低血液中膽固醇與三酸甘油脂含量。經過餵食8週的實驗性飼料後,在成長方面除肝體比(Hepatosomatic index, HSI)以攝食6.0% 青海菜粉組較高外,其他指數皆無顯著差異,但活存率方面以6.0%及9.0% 青海菜粉組較低,各為69%及75%。而在所有飼料組中日本鰻之血脂方面,攝食3.0%及9.0%青海菜粉組能明顯降低血清中膽固醇含量;攝食3.0%、6.0%及9.0%青海菜粉組明顯降低血清中三酸甘油脂含量。 在肝臟中GSH (Glutathione)含量與抗氧化酵素 GSH Rd (Glutathione reductase)活性方面,各飼料組皆無顯著性差異。在CAT (Catalase)方面,攝食3.0%以上的青海菜粉能有效提升其活性。在GST (Glutathione S-transferase)方面,攝食3.0%青海菜粉組較其他組有較高的活性。在GSH-Px (Glutathioine peroxidase)方面,攝食6.0%及9.0%青海菜粉組較控制組有較高的活性。在SOD (Sueroxide dismutase)方面,攝食.0%及9.0%青海菜粉組獲得較高的活性,而以攝食3.0% 青海菜粉為最低。 在Cd2+的急性染毒(0.69 mg/L, 48 hrs)下,與跟其他飼料組相較,攝食6.0%與9.0%青海菜粉組有較高的CAT活性;攝食9.0%青海菜粉組獲得最高的GST活性;攝食6.0%與9.0%青海菜粉組有較高的SOD活性。飼料中添加6.0%與9.0%的青海菜粉組可以有效的增加日本鰻抗氧化能力,同時在急性染毒的情況下,可以有效的清除體內過多的自由基,保護細胞降低傷害。

The parameters such as growth performance, liver antioxidant enzyme activity, liver detoxifying enzyme activity, and serum cholesterol content and triglyceride content were evauated when Japanese eel (Anguilla japonica) were fed diets containing Monostroma nitidum powder with 0%, 3.0%, 6.0%, and 9.0% added into feed as tested groups during 8 weeks. Results indicated that eel fed a diet of 6.0% group for the growth performance, had significant high HSI (Hepatosomatic index) but with no significant differences among the other parameters. The survival of eel that fed a diet of 6.0% and 9.0% groups was low at 69% and 75%, respectively. For eels fed diets of 3.0% and 9.0% groups showed a significant decrease in their serum cholesterol content. A significant reduction of the serum triglyceride content was observed for diets of 3.0%, 6.0%, and 9.0% groups. The results marked that GSH (Glutathione) content and antioxidative enzyme activity of GSH Rd (Glutathione reductase) had no significantly difference among all tested groups. For CAT (Catalase) activity, those fed 3.0%, 6.0% and 9.0% groups exhibited a significant higher activity than that of 0 % group. GST (Glutathione S-transferase) activity for eels fed 3.0% group was significantly higher than other groups. GSH-Px (Glutathioine peroxidase) activity for 6.0% and 9.0% groups were significantly higher than that of 0% group. SOD (Sueroxide dismutase) activity for 0% and 9.0% groups were significantly higher than other groups. The lowest SOD value was found for eels fed 3.0% group. After Cd2+ acute toxication (0.69 mg/L, 48 hrs), CAT activity for eels fed 6.0% and 9.0% groups had significantly higher value than other groups. The highest value of GST activity was observed for eels fed 9.0% group. SOD activity for eels fed 6.0% and 9.0% were significantly higher than other groups. For groups added of 6.0% and 9.0% Monostroma nitidum powder would significantly increase the antioxidant activity of Japanese eel. In addition, under acute toxication, it may effectively eliminate the superfluous free radicals and protect cell from injury in eel.
