中草藥抗氧化能力之探討

Antioxidant activity of selected Chinese herbal extracts

摘要


近年來,隨著國人健康意識逐漸抬頭,中草藥植物的開發與運用,已成為世界性的發展趨勢。本研究的目的在於評估五種不同之中草藥,分別為龍膽草(Gintiana)、手掌參(Gymnadenia conopsea)、天山雪蓮(Smallanthus sonchifolius)、石榴根(Punica granatum)、針層孔菌(Phellinus igniarius)以不同極性溶劑萃取其抗氧化能力之探討。經實驗結果顯示;於清除DPPH自由基能力上,以乙醇萃取之石榴根萃取液為最佳,可清除90.53%之DPPH自由基;於螯合亞鐵能力評估上,其乙醇萃取液、乙酸乙脂萃取液、水萃取液均可達到良好的清除能力,至少可達70.66%清除力;於清除超氧陰離子部分,以乙醇萃取之針層孔菌萃取液達到最好清除效果,可清除99.54%之超氧陰離子;於還原力方面,則以乙醇萃取之龍膽草萃取液效果最佳,可達73.83%還原力;而於類黃酮測定方面,以乙醇萃取之龍膽草萃取液、水萃取之龍膽草萃取液、石榴根萃取液及天山雪蓮萃取液效果最佳,其類黃酮含量分別為98.00%、99.50%、90.00%、99.45%;於總酚化合物方面,以水萃取之龍膽草、石榴根之含量較佳。本研究將針對龍膽草進一步分析處理,經實驗結果顯示:於螯合亞鐵能力進行評估,當濃度達300 μg/mL,其乙醇萃取螯合能力可達96.80%;於清除超氧陰離子部份,當濃度達300 μg/mL,其水萃取液可清除94.30%之超氧陰離子;於H2O2方面,當濃度達50 μg/mL,其乙醇萃取液可清除75.01% 之H2O2;而針對總抗氧化物ABTS+而言,當其濃度達300 μg/mL,其水萃取液及乙醇萃取液可達100%。藉此可推測,當龍膽草萃取液達一定濃度時,其具有良好之抗氧化能力。本研究進而利用管柱層析純化分離,將部分純化物質重新評估其抗氧化活性。實驗結果顯示,龍膽草水萃取液經由協同作用,可有效提升螯合亞鐵離子能力及清除超氧陰離子能力。

In recent years, the health consciousness is gradually accepted by consumers. Therefore, the development and application of the Chinese herbals, have already become the world trend and more attractive to industries. The objective of current study is focus on assessing the antioxidant activity of selected Chinese herbals, Gintiana, Gymnadenia conopsea , Smallanthus sonchifolius , Punica granatum Linn , and Phellinus igniarius extracted by various solvent. The results indicated that 90.53% DPPH free radical scavenging effect was exhibited when ethanol extract of Punica granatum Linn was employed. For chelating ability, more than 70% ferrous ions capture can be shown with solvent-independent extract of Punica granatum under specified conditions. Besides, 99.54% superoxide anion scavenging activity was observed, when Phellinus igniarius ethanol extract was used. Furthermore, the best reducing capacity, 73.83%, was found if ethanol extract from Gintiana was employed in the present study. The measured flavonoid contents of Gintiana ethanol extract, Gintiana water extract, Punica granatum Linn water extract and Smallanthus sonchifolius water extract are 98.00%, 99.50%, 90.00% and 99.45%, respectively. The higher contents of total phenol contents were observed in water extract from Gintiana, Punica granatum Linn, and snowfield grass. Moreover, the advanced study on Gintiana was conducted due to its antiinflammatory property and the results demonstrated that more than 96% of chelating ability was measured when 300 μg/mL ethanol extract was used. The water extract of Gintiana showed 94.03% superoxide anion scavenging activity if up to 300 μg/mL concentration was employed. For H2O2, more than 75% scavenging capacity was observed when 50 μg/mL of ethanol extract was used. The ABTS+ scavenging activity were almost reached 100% with dose-independent of water and ethanol extract of Gintiana. In conclusion, the antioxidant activity has positive relationship with total phenolic contents under specified conditions. Furthermore, partial purified active substances of Gintiana for antioxidant activity identification will be conducted. The results demonstrated that significantly synergistic effects were observed in ferrous ion chelating ability and superoxide anion scavenging activity when different chromatographic fractions were employed.
