以免疫測定法試驗免疫促進劑對脂多醣之吸附效果以及白蝦飼料中添加β-葡聚醣、蒙脫石與海藻酸鈉對副溶血性弧菌攻毒之保護效果

摘要


以免疫測定法試驗免疫促進劑β-葡聚醣(βG、蒙脫石(MMT)與海藻酸鈉(SA)對脂多醣(LPS)之吸附效果。顯示最佳吸附濃度分別為βG 1000 μg/mL、βG 500 μg/mL、βG 250 μg/mL、MMT 500 μg/mL與SA 500 μg/mL對LPS 2.5 μg/mL之組合。評估白蝦飼料中添加βG、MMT與SA對副溶血性弧菌攻毒之生長影響與保護效果,以五組(βG、MMT、SA、陰性與陽性對照)各三重複每組10尾白蝦進行試驗,白蝦在15日馴化後,飼料中添加免疫促進劑βG、MMT與SA 各30日之後,以副溶血性弧菌(Vibrio parahaemolyticus)攻毒結果顯示MMT之20 g/kg組之飼料效率1.55 ± 16.21g優於對照組的2.07 ± 7.56g(p < 0.05。在增重方面,同樣是MMT的20 g/kg組之增重3.24 ± 0.32 g優於對照組的2.44 ± 0.32 g(p < 0.05)。三個免疫促進劑組的存活率為 98%皆高於陽性對照的50%。在組織病理學結果,Vibrio parahaemolyticus (Vph) 攻毒後都有嗜酸性球的增加。MMT組(20 g/kg組)有較好的表現,綜合顯示三個免疫促進劑對LPS皆有吸附效果,在飼料效率、生長效力、存活率、腸道細菌與各免疫參數之研究(如:血細胞總數與抗細菌力)皆能幫助白蝦對抗LPS與Vph感染試驗之影響。

The immunostimulants of β-Glucan (βG), Montmorillonite (MMT) and Sodium Alginate (SA) were used to binding with lipopolysaccharide (LPS) test by enzyme-linked immunosorbent assay (ELISA) method. The results indicated that the optimal bound concentrations were shown 1000, 500 and 250 μg/mL of βG bound with 10 μg/mL of LPS, 500 μg/mL of MMT bound with 1.25 μg/mL of LPS, and 500 μg/mL of SA bound with 2.5 μg/mL of LPS, respectively. The study of dietary βG, MMT and SA on the growth and survival rate for challenged with Vibrio parahaemolyticus (Vph) in Litopenaeus vannamei (Lv) were assessed. Collect of healthy shrimps were cultivate for grouping with ten shrimps each tank in triplicated (negative control, positive control and treatment groups). The treated Lv showed significant different (P < 0.05) improvement lower of feed conversion ratio (FCR) D1 (MMT 20 g/kg) 1.55 ± 16.21 compared to negative control Lv 2.07 ± 7.56. Higher of weight gain in D1 (MMT 20 g/kg) 3.24 ± 0.32 g compared to negative control Lv 2.44 ± 0.32 g significant different (P < 0.05). Survival rate (98%) of βG, MMT and SA treated groups were shown significant different (P < 0.05) compared to positive control Lv (50%) at 5% significant level. The Vph challenged experiment resulted in 50% mortality in control positive groups. The Lv of treated with immunostimulants were totally protected with resulted in 95 - 98% survival against Vph and examined by polymerase chain reaction method. In histopathology study, most of symptoms because of challenged by Vph infection was increased level of eosinophil cells. In addition, MMT group was significantly better protection results based on all parameters. The most highest protective ability among immunostimulants fed group was D1 (MMT 20 g/kg) compared with the other groups. In brief, the results of this study were described that dietary immunostimulants of βG, MMT and SA successfully progresses their bound with LPS, feed conversion ratio, growth rate, survival rate and immune parameters study (total hemocyte count and bactericidal activity) could help secures Lv against LPS and Vph challenged
