豨薟草萃取物之抗糖化生化活性與抗氧化能力探討

Antiglycation and antioxidant effects of Siegesbeckia orientalis extracts

摘要


豨薟草屬雙子葉菊科草本植物,具有治療四肢麻痺、風濕關節痛、筋骨疼痛、腰膝無力、高血壓、疔瘡腫毒、外傷出血等功效,但至今文獻上皆無報導豨薟草之抗糖化作用。本研究將豨薟乙醇萃取物(Siegesbeckia orientals extract, SE)以不同溶劑進行分層萃取,取得正己烷分層萃取物(SE-Hex)、乙酸乙酯分層萃取物(SE-EA)和甲醇分層萃取物(SE-MeOH),並評估不同分層萃取物之抗氧化與抗糖化活性。研究結果顯示,總多酚及類黃酮含量皆以SE-EA含量最多,其數值分別為176.9 mg catechin/g sample及5.3 mg catechin/g sample;抗氧化活性測試中,SE-EA的DPPH、ABTS自由基清除活性及還原力效果最佳。此外,在抗糖化活性測試中,也以SE-EA對抑制糖化終產物(AGEs)的生成、α-澱粉酶(IC50值為362.3 μg/ml)及α-葡萄糖苷酶(IC50值為119.0 μg/ml)的活性最佳,並呈現明顯的劑量依存性。在體外實驗結果顯示,豨薟草萃取物不僅具有成為糖尿病輔助食品或藥物之潛力,也可用於治療或預防糖尿病併發症的發生。

Siegesbeckia orientalis linne, which belongs to Asteraceae, is an herbal and dicotyledonous plant. It is a traditional Chinese medicinal herb endowing with the medicinal facts to treat limb paralysis, rheumatism, joint pain, aching pain, lassitude, weakness, hypertension, detumescence, detoxification, trauma, bleeding. However, no literature reports its anti-glycation activity so far. In this study, S. orientailis was firstly extracted by ethanol. This SE extract was then extracted and partitiond sequentially with hexane, ethyl acetate (EA) and methanol, and their extracts were named SE-Hex, SE-EA and SE-MeOH, respectively. The antioxidant and anti-glycation effects of these extracts were investigated. The experimental results showed that the toal phenolic content (176.9 mg gallic acid/g sample) and the total flavonoids content (5.3 mg catechin/g sample) of SE-EA was the highest among these extracts. The SE-EA has the most significant antioxidant activities, including DPPH radical-scavenging capacity, ABTS radical-scavenging capacity and reducing power. For anti-glycation activities, SE-EA also showed the best results in the inhibition of advanced glycated end products (AGEs), and inhibitory activities against α-glucosidase (IC50 = 362.3 μg/ml) and α-amylase (IC50 = 119.0 μg/ml). These in vitro studies indicate that Siegesbeckia orientalis linne may be effective in preventing and treating diabetes mellitus and diabetic complications.
