牛樟芝為台灣特有種真菌,別稱樟芝或牛樟菇。牛樟芝主要的生理活性成分為三萜類(Triterpenoids),它是由30個碳所結合而成六角形或五角形結構形成的天然有機化合物的統稱。三萜類在自然界分佈廣泛,部分的植物、動物及菌類都含有此類化合物,但從不同動植物身上所發現的三萜類種類及含量又有些許不同,三萜類具有抑制癌細胞生長和抗氧化活性的特性,一般的三萜類多是由羊毛甾醇(Lanosterol)或麥角甾醇(Ergosterol)所組成,而牛樟芝子實體具較高藥理活性之三萜類大都以麥角甾醇為主,其另含有維生素、多醣體、超氧歧化酵素、腺苷、免疫球蛋白及其他微量元素。而本實驗運用HPLC定量分析牛樟芝三萜類,初步實驗證實內在標準品(S)與三萜類可以相同的溶劑沖提,且兩者在HPLC當中有著不同的滯留時間,所以內在標準品(S)適合做為三萜類HPLC定量分析的內在標準品化合物,適當發展之後從HPLC分析圖譜的層析鋒面積比值計算與三萜類濃度之間的相關性,依其良好的相關性即可建立標準曲線並由標準曲線可推算出每一種樟芝樣品溶液中所含三萜類的濃度,並比較瓶培樣品之間的差異性。
Antrodia cinnamomea (AC) is a unique fungus in Taiwan, also known as Chang-chin or niu-chang-ku. The major bioactive constituents of AC are triterpenoids compound, which is 30 carbons combination of pentagonal or hexagonal structure formed of natural organic compounds collectively. triterpenoids are widely distributed in nature such as: plants, animals, and fungi. The configuration and contents of triterpenes were varied in different sources. Triterpenoids may possess a cancer cell growth inhibited and anti-oxidation activity characteristics. Chemically, the predominated form of triterpenoids was found to be ergosterol-based structure, such as: lanosterol or ergosterol in fruiting bodies of AC. Furthermore, the high pharmacological activity constituents namely, vitamins, polysaccharides, superoxide dismutase, adenosine, immunoglobulins, and other trace elements were also found in this fruiting body. The present study was designed to use HPLC quantitative analysis AC triterpenoids. Preliminary results demonstrated that the selected internal standard and target triterpenoids may be the same solvent punch mention, but have different retention time in HPLC chromatogram. Therefore, this internal standard is available for triterpenoids quantitatively analyzed by HPLC. After proper development from HPLC analysis correlation wave front area ratio, the calculated concentration profiles of triterpenoids could be used to create a standard curve. The concentration of each triterpenoid contained in the solution of AC sample could be deduced from this standard curve, and compare the differences in AC bottle culture samples.
