葡萄糖胺衍生物萃取與生醫材料改質應用

摘要


本篇目的是利用動物副產物供分離葡萄糖胺類物質,將價值較低的副產物萃取 使其成為有高經濟價值的物質。主要分為三個部分,第一部分為從雞冠萃取出玻尿 酸,目前連續萃取所得的產率分別為:第一次萃取0.50% ,第二次萃取0.63%,第 三次萃取0.13% ,玻尿酸純度則以HPLC 測試。第二部分為利用蝦殼粉萃取出幾丁 質及幾丁聚糖,結果利用100g 的蝦殼粉可獲得6.91g 的幾丁質,產率為6.91%。第 三部分則是利用猪耳朵、猪氣管來萃取軟骨素,其產率分別為7.15%與0.70%。 萃取完之後,接下來利用所得到的產物進行改質實驗,期望能在盡量不改變原 本材料性質的前提下,另外再附加上其他不同的性質,像是增加抗菌性、提昇細胞 生長或是促進凝血等等優點。由於直接以葡萄糖胺(含幾丁質、幾丁聚醣)作為手術 縫線,常因為縫線機械強度不足與耐酸性不佳的問題,在臨床上並未大量使用。我 們將生物可分解縫線經由電漿表面改質處理,浸泡於幾丁聚醣溶液並以UV 光照 射,使幾丁聚醣接枝上縫線表面,最後再測試縫線是否具有抗菌能力,從實驗的結 果得知,改質接枝後的幾丁聚醣確實可增加縫線本身的抗菌效果。另外我們也試著 將所萃取得到的軟骨素接枝至生物高分子孔洞薄膜上(聚羥基丁酯戊酯;PHBV),並 與未接枝薄膜、接枝明膠的薄膜進行比較,結果軟骨素薄膜所培養的細胞數明顯超 越其他兩組,證實軟骨素接枝於高分子薄膜表面可以提高細胞貼附的效率。

This thesis developed the extraction methods for extracting glucosamine from animal by products. Hyaluronic acid was extracted from the comb of roosters. The goal is to enhance production rate as well as the purity of hyaluronic acid. The production rate of hyaluronic acid was 0.50% for the first extraction, 0.63% for the second extraction, and 0.13% for the third extraction. HPLC was used to determine the purity of the hyaluronic acid. Chitin and chitosan from shrimp shell was another source of glucosamine. The production yield was 6.91% when extracted with alkaline solution. Chondroitin from pig ears and pig trachea was obtained with acetic acid extraction process. The production yield from pig ears is 7.15% while the production yield from pig trachea is .07%. The glucosamines obtained from animal were used in modification experiments. We used the Plasma treatment method to graft glucosamine onto different materials, including pHBV membrane and sutures. The cell proliferation and adherence of pHBV (poly(3-hydroxybutyrate-co-3-hydroxyvalerate))membrane also can be further improved by plasma treatment. Biodegradable sutures, Polyglactin, grafted with chitosan were tested to determine it’s properties. The results indicate that modified sutures has better antiseptic property than unmodified sutures.
