利用冷電漿及接枝技術表面固定厚朴酚

Surface Immobilization of Magnolol by Cold Plasma and Graft Polymerization

摘要


厚朴酚(Magnolol)具有抑制細胞增生之藥物,亦是東方傳統中藥材,醫療上用以作為治療胃部疾病,心血管和過敏症的疾病並有抗焦慮、抗氧化、抗癌…等功效。本研究利用表面改質技術希望能將其固定於材料表面,以供在生醫材料之應用。在本研究中,利用電漿輔助化學氣相沉積於材料表層披覆一有機矽化物或有機錫化物薄膜作為無機基材表面中介層,使其表面帶自由基源,並以四種方式固定:(1) 直接固定Magnolol (2) UV光接枝 (Alginate/ Magnolol)複合膠體 (3) UV光接枝固定 (Acryl acid/ Magnolol) (4) 先熱接枝 (Acryl acid/ Magnolol),再以褐藻酸鈉(Alginate)與鈣交聯成含藥載體。期待製作具生物適應性又有因Alginate分解而藥物釋放作用的水膠表面。經由FTIR、QCM…等分析確定經電漿沉積後以上方法都可以接枝固定Magnolol但是量很少,其中以接枝AAc混合Magnolol或Alginate混合Magnolol都可成功固定含Magnolol複合物在基材表面。初步藉由平滑肌細胞,肝癌細胞及內皮細胞培養的結果可得知有Magnolol 成分表現出有抑制細胞生長的效果。

Magnolol, is isolated from the root and stems bark of Magnolia offcinalis, which has been a medicine in traditional east country for a long time. It has multiple pharmacological effects, such as antidepressant, anticlastogenic, antioxidant, antithrombotic, antimicrobial, antiallergic, and antifungal. In this study, Magnolol was used to immobilize on surface of materials by surface modified technology for biomaterial application. By using plasma method deposits organic silicone compound films or organic stannum compound films for the inter layer of inorganic surface, in order to created peroxide on the surface. And then use UV induced surface graft polymerization to graft (Alginate/Magnolol) composite gel or Acrylic acid (AAc) mixed Magnolol then cross linking with Alginate and Calcium Chloride to form a drug carrier on polymer coated surface. Through FTIR、QCM…analyses could demonstrate that after plasma deposition can UV-graft Magnolol, but the amount is about 4.76Χ10-4μg/cm2. Therefore graft of mixing AAc/Magnolol and Alginate/Magnolol solution can immobilize Magnolol complex on the surface of substrates successfully.
