人工植牙後的植體與齒槽骨的骨整合需要三至四個月的時間,因此本研究擬利用超音波刺激以加速其作用。實驗分三個部分:細胞培養、器官培養與動物實驗。研究將評估超音波刺激細胞、器官與動物體之反應與活性,並觀察超音波是否促進骨整合作用。 在細胞培養實驗中,將類骨母細胞植覆在鈦金屬上,使用不同參數的超音波刺激,結果顯示脈衝模式的超音波刺激在細胞存活率和細胞死亡率以及細胞週期都具有優異表現;體外的器官培養是將活體小鼠取出的頭蓋骨挖洞和植體一起培養並且接受超音波刺激;結果器官培養發現脈衝模式的0.15W/cm2骨礦化和骨引導是脈衝組中最突出的。最後將植體以手術方式植入兔體脛骨,組織染色和超音波影像也顯示出脈衝模式的0.15W/cm2是較佳的參數。
The process of osseointegration in dental implant and alveolus will take about 3 to 4 months. In this study, the effect of ultrasound stimulation was utilized to accelerate the osseointegration in dental implant and alveolus. The cell culture, organ culture and animal study were evaluated under several ultrasound stimulations. The results of cell culture demonstrated that all the pulse-wave ultrasound stimulation had better performance than continuous ultrasound stimulation in cell survival and cell cycle. The 1:4 pulse-wave, 0.15W/cm2 group had the superior effects among all other ultrasound stimulation. Further, the histological test also showed the similar results in animal study.
