槲皮素對腦缺血大鼠之大腦皮質脂質過氧化作用、抗氧化酵素活性與微量元素之效應

摘要


根據衛生福利部統計,腦中風在台灣人口十大死因中排行第四名。研究顯示,腦中風會造成腦內自由基濃度上升,從而造成大腦氧化性壓力的產生並導致氧化性損傷。先前的研究指出,適量的微量元素以及抗氧化酵素能有效的保護大腦,減少活性氧化物造成的氧化性損害。槲皮素屬於黃酮類化合物,主要存在於許多蔬菜水果例如:洋蔥與蘋果等食物中。槲皮素具有抗發炎、抗過敏、抗腫瘤、抗氧化以及神經保護作用。目前為止槲皮素針對腦缺血大腦的微量元素、抗氧化酵素與脂質過氧化作用的影響還不清楚。實驗結果顯示,腦缺血前給予槲皮素能夠有效的增加抗氧化酵素、微量元素鎂 (Mg)、鋅 (Zn)、硒 (Se)的濃度;此外,槲皮素也能顯著減少脂質過氧化作用與必需元素鐵 (Fe)、銅 (Cu)以及有毒金屬鉛 (Pb)、砷 (As)、鉻 (Cr)、鋁 (Al)的濃度。綜合所有結果可以得出結論,槲皮素的神經保護機制與降低脂質過氧化狀態,提高抗氧化酶活性和改變缺血性腦皮質中的微量元素濃度密切相關。

According to the report from the Ministry of Health and Welfare, strokes ranked fourth among the top ten causes of death in Taiwan. Studies have shown that stroke not only generates toxic free radicals but also results in further oxidative stress and brain damage. Previous studies have pointed out that proper trace element level and antioxidant enzyme activity are required for the brain for maintaining their normal metabolism. Quercetin is a flavonoid compound mainly existed in many fruits and vegetables such as onions and apples. Previous investigation has revealed the fact that quercetin possessing a variety of beneficial functions such as anti-inflammation, anti-allergic, anti-tumor, anti-oxidation and neuroprotection. So far, quercetin has not been fully discussed about its influence on the changes of trace elements, antioxidant activity and lipid peroxidation during cerebral ischemic insult. Our experimental results show that administration of rats with quercetin prior to cerebral ischemia can effectively increase antioxidant activity, and trace element level of magnesium (Mg), zinc (Zn), and selenium (Se). Furthermore, reduced lipid peroxidation intensity and declined trace element concentration of iron (Fe), copper (Cu), lead (Pb), arsenic (As), chromium (Cr), and aluminum (Al) was found. Taking together, it seems possible that neuroprotective mechanism of quercetin is closely associated with declining lipid peroxidation status, elevating antioxidant enzyme activity, and altering trace element level.
