許多研究發現多種蔥屬(Allium)蔬菜具有優良之抑菌效果,抑菌範圍包括許多腐敗菌及腸內病原菌,本研究探討省產韭菜(Allium odorum Linn.)及寬葉韭(Allium hookeri Thwaites)之抑菌作用及抗氧化作用。韭菜及寬葉韭經採集鑑定後,以95%的乙醇萃取,濃縮乾燥後分別以正己烷、乙酸乙酯、正丁醇乙醇及水等不同極性之溶劑分層萃取,然後再進行抑菌圈試驗。選用的菌株包括大腸桿菌(Escherichia coli)、鼠傷寒沙門氏菌(Salmonella typhimurium)、金黃色葡萄球菌(Staphylococcus aureus)、仙人掌桿菌(Bacillus cereus)等。 實驗結果顯示,韭菜及寬葉韭乙酸乙酯層萃取物的抑菌效果最佳且對各試驗菌株皆有抑菌效果,而又以韭菜的抑菌效果優於寬葉韭;水層及正己烷萃取物在本實驗採用之濃度(0.2g/mL)下並無抑菌作用。從EtBr(溴化乙錠)染色觀察結果推論,韭菜及寬葉韭具殺菌作用,因而EtBr 可通過細胞膜與核酸結合,呈現較佳之染色效果。 在抗氧化作用方面,DPPH自由基清除能力皆以正丁醇層效果最佳,其中韭菜效果又優於寬葉韭;而亞鐵離子螯合能力,則以乙醇、正丁醇及水層具螯合效果;還原力部分則以乙酸乙酯層效果最佳,其次為正丁醇層。
The purpose of this research was to evaluate the antibacterial and antioxidative activities of Chinese leek. Allium odorum Linn. and Allium hookeri Thwaites were extracted with three times of ethyl alcohol. The extracts was then partitioned by n-hexane, ethyl acetate, n-butyl alcohol and water. Experimental results indicated that n-hexane and water extracts exhibited no significantly antimicrobial activity. The ethyl acetate extracts especially presented the best antimicrobial activity, and A. odorum L. better than A. hookeri Thwaites. From the results of Ethidium bromide stained, it showed that the dye could cross the broken cell membrane of these bacteria treated with these extracts. In the analysis of DPPH scavenging activity, n-butyl alcohol extracts exhibited the hightest scavenging activity especially the A. odorum L..Ethyl alcohol, n-butyl alcohol and water extracts in A. odorum L. could present the chelating activities. The ethyl acetate extracts showed the greatest reducing power.
