河車大造丸對兩側卵巢切除大鼠骨質效用之研究

Effect of Ho-Tr-Da-Za-Wa on Bone Matrix in Rats with Bilateral Ovariectomy

摘要


目的:停經後婦女容易造成骨質疏鬆症,一般認為與體內鈣的代謝和停經後雌激素的減少有密切關係。雖然雌激素可改善停經後的骨質疏鬆症,但會增加子宮內膜癌或乳癌的發生率。中醫認為停經後婦女的骨質疏鬆症,大部分是由於腎陰虛所導致,因此本研究的目的是探討具有補腎陰作用的河車大造丸對兩側卵巢切除大鼠的骨量效應。 方法:將35隻重量200-250公克、90天鼠齡之雌性Sprague-Dawley(SD)大鼠,隨機分成每7隻一組如下:1)假性卵巢切除組(SH):將老鼠兩側的卵巢取出放置於腹腔外,然後立即再放回原位,但不切除;2)控制組(Control):不施行任何手術或投予任何藥物。3)卵巢切除組(OV):將老鼠兩側的卵巢切除;4)卵巢切除加河車大造丸組(OV+D):將老鼠兩側卵巢切除28天後,第29天開始口服河車大造丸至第56天;5)假性卵巢切除加河車大造丸組(SH+D):同SH組,但投予河車大造丸,方法同(OV+D)組;手術後第57天將大鼠犧牲,取出兩側脛骨,施行接觸顯微放射攝影觀察骨質密度。 結果:(OV+D)組的脛骨骨質密度比OV組高。 結論:河車大造丸能增加兩側卵巢切除SD大鼠脛骨的骨質密度,推論河車能使用於停經後婦女的骨質疏鬆症。

Objectives: The etiology of osteoporosis in the women with menopause is resulting from the imbalance of calcium metabolism and the decrease of estrogen has been reported. Through estrogen can improve menopause osteoporosis, it also can increase the incidence of endometrial and breast cancer. According to the theory of Traditional Chinese Medicine, the most etiology of the osteoporosis in women with menopause is due to deficiency of kidney-yin, therefore the purpose of the present study is to investigate Ho-Tr-Da-Za-Wa that has the effect of supplementing kidney on bone matrix in rats with bilateral ovariectomy. Mehtods: a total of 35 female Sprague-Dawely (SD) rats, weight 200-250 g, 90 days in age, was randomly divided into 5 groups of 7 rats as follows: 1) sham group (SH), exposing the bilateral ovary to outside of abdomen, and then recovery immediately to original site, but not excised; 2) control group (Control), no any operation or any drug was administer; 3) Ovary excising groups (OV), bilateral ovary of the rats were excised; 4)Ovary excising plus Ho-Tr-Da-Za-Wa groups (OV+D), the method identical to the OV group, but Ho-Tr-Da-Za-Wa was oral administered from 29(superscript th) day to 56(superscript th) day after operation; 5) sham plus Ho-Tr-Da-Za-Wa group (SH+D), the method identical to the SH group, but Ho-Tr-Da-Za-Wa was oral administered from 29(superscript th) day to 56(superscript th) day after operation. The rats were sacrificed at the 57(superscript th) after operation, and their tibia bones were took out. The density of bone matrix was measured by using contact microscopic photographs computer system. Results: The density of tibia bone matrix was greater in the OV+D group than OV group in rats with bilateral variectomy. Conclusion: Ho-Tr-Da-Za-Wa can increase the density of tibia bone matrixt in rats with bilateral ovariectomy, suggesting Ho-Tr-Da-Za-Wa can treat osteoporosis in women with menopause.
