運用都卜勒超音波驗證牡丹皮萃取物降主動脈阻力特性

Effect of Extracts of Root Cortex of Paeonia Suffruticosa on Reducing Aortic Resistance in Rats by Doppler Echocardiography

摘要


牡丹皮(Root cortex of Paeonia、suffruticosa)爲牡丹(Paeonia suffruticosa Andr.)乾燥之根皮,依記載具抑制血小板凝集、抗菌、抗發炎等效果。本實驗將以超臨界二氧化碳(supercritical carbon dioxide)針對牡丹酚進行最佳萃取條件建立;再以心臟都卜勒超音波(Echocardiography Doppler)驗證其萃取物之降血壓特性。 實驗發現當超臨界二氧化碳萃取壓力爲工100atm,溫度60℃,萃取時間30分鐘下可得牡丹酚最佳萃取效率,若於萃取過程中添加萃取槽體積之13.6%的甲醇,對於萃取效率有顯著的增加。動物降血壓測試方面發現給予劑量1.41mg/kg,其血壓下降32.0±4mmHg,以心臟都卜勒超音波觀察其主動脈阻力指數,可發現動脈阻力指數(Aortic Resistance Index, ARI)比對照組下降至約83.8±0.7%;其持續時間爲376.0±21秒。 利用心臟都卜勒超音波以非侵襲方式測量大白鼠主動脈阻力指數,將有助於觀察降血壓功效之中草藥對大白鼠血壓的影響。

Root cortex of Paeonia suffruticosa, also called ”Mudanpi”, has been found that can promote blood flow to remove blood stasis, resist inflammation, and reduce blood pressure. In this study, we established the method for supercritical carbon dioxide extraction in root cortex of Paeonia Suffruticosa and study the effect on reducing aortic resistance by animal experiment, which assayed the aortic resistance by Echocardiography Doppler. In our preliminary data, the best extraction efficiency is the pressure at 100atm, temperature at 60℃, 13.6% methanol as modifier and extraction time in 30 minutes. The rats under general anesthesia were cannulated from right femoral arteries with polyethylene tubing (PE50) to monitor physiological parameters. The relative percentage of aortic resistance index (ART) after injection of extracts was lower than the value after injection of vehicle (0.9% NaCl). We found the aortic resistance index (ART) could reduce 20% after administration of extracts of Paeonia suffruticosa (1.41mg/kg of body weight). The duration time of action is 376±21 s. These observations have led to the suggestion that the crude extracts from supercritical carbon dioxide extraction act through the reduction of aortic resistance to decrease blood pressure response.
