前處理缺氧及後處理缺氧在中大腦動脈阻斷的中風模式其低氧誘導因子(HIF-1α)、基質金屬蛋白質酶2(MMP-2)及熱休克蛋白質70(HSP70)在肺的表現

摘要


臨床上因血栓、失血或組織病變引起組織細胞缺氧的情形極爲常見,缺氧後不僅影響一般血管新生及組織的修復,也極易誘發腫瘤之惡化及轉移。在本研究中將利用溫和的低氧前處理或後處理,來探討其對活體織抗壓潛能之影響,所使用之中風缺血模式係以大白鼠阻斷中大腦動脈手術,並在中風前後分別依組別給予過和的低氧前處理或後處理(12% O2 pre-hypoxia or post-hypoxia)8小時,在動物犧牲後取出與缺氧代償密切相關之肺臟組織作均質處埋,以西方轉漬法(western bloting)分別檢測低氧誘導因子-1α(Hypoxic-Inducible factor-1α; HIF-1α)、基質金屬蛋白酶2(Matrix Metalloproteinases-2; MMP2)及熱休克蛋白質70(Heat Shock Protein 70; HSP70)表現量。近年有許多研究指出織織缺氧後即有HIF-1α(HIF-1α)蛋白表現增加,在此研究中將被視爲缺氧及中風後對肺臟血管增生的指標。而MMP2的表現將會造成織織結構之重建、與組織的病理變化密切相關,在組織病理狀態時組織細胞外基質會被此MMP2分解。HSP70爲一種有效的保護性蛋白質,其表現量可作爲評估低氧處理對肺臟組織所產生之保護效應。本實驗結果顯示:HIF-1α的表現在低氧處理後均會增加(無論是在前處理組或後處理組),而中大腦動脈阻斷所引起的傷害會看到MMP2的表現增加。低氧前處理會誘導HSP70的表現是可以保護中大腦動脈所引起的傷害。且HSP70的表現是可能和MMP2、HIF有些關聯。此數據將提供我們日後作低氧前處理或後處理中風後,肺組織進行磁共振造影的影像比對。

Cellular hypoxia induced by bleeding, thrombus or tissue degeneration is common in clinic. The most impressive of those responses is the increased tissue oxygen delivery. Another response is the increased metastasis in tumor. Several studies have demonstrated that a period of pre-hypoxic or post-hypoxic can provide protection against a subsequent severe insult. In the present report, we study the effect of hypoxic pretreatment or postreatment in stroke induced by occlusion of middle cerebral artery occlusion on the expression of Hypoxia Inducible Factor-1α(HIP-1α), Matrix Metalloprotcinase-2(MMP2), Heat Shock Protein 70(HSP7O). Health adult rats were divided five groups including control. ischemia without post hypoxic treatment, prehypoxic ischemia, post hypoxic ischemia, and ischemia without prehypoxic treatment. The ischemic brain model was conducted by occlusion of middle cerebral artery. When the stroke was induced, the rats were pretreated or postreated into hypoxic chamber (12% O2). We detected the relationship of HIF-1α, MMP2, HSP7O in lung. The results will provide some hints in magnetic resonance imaging (MRI) data.
