醋酸纖維素/紫草素奈米纖維膜之製備及生物相容性分析

摘要


紫草素(shikonin)是一種傳統中草藥,從紫草科植物紫草的萃取物中分離得到,具有多種藥理活性,如促進傷口癒合、抗菌、抗發炎、抗氧化、抗病毒及抗腫瘤等活性。本研究將醋酸纖維素包覆紫草素,通過靜電紡絲加工技術製備奈米纖維膜,應用於傷口敷料領域。並探討醋酸纖維素/紫草素奈米纖維膜的物化性質及生物相容性。採用掃描式電子顯微鏡(SEM)觀察醋酸纖維素和醋酸纖維素/紫草素奈米纖維膜的表面形態,其纖維平均直徑約在1300 nm到1330 nm之間。實驗結果表明醋酸纖維素奈米纖維膜具有良好的平衡含水率及水蒸氣穿透率。抗氧化活性則採用DPPH自由基清除能力方法評估。經由抑菌圈實驗顯示醋酸纖維素/紫草素奈米纖維膜對於大腸桿菌和金黃色葡萄菌具有良好的抗菌活性。體外間接MTT分析方法表明醋酸纖維素/紫草素奈米纖維膜對L929小鼠纖維母細胞具有良好的細胞相容性。綜合以上結果,醋酸纖維素/紫草素奈米纖維膜具有應用於抗菌敷料的潛力。

Shikonin, a traditional Chinese herbal medicine made from the extracts of the plant Lithgospermum erythrorhizon, exhibits various pharmacological activities, such as wound healing, antimicrobial, anti-inflammatory, antioxidant, antiviral, and antitumor activity. The aim of this study was to develop and fabricate cellulose acetate (CA) nanofiber mats containing shikonin via electrospinning technique for wound dressing applications. The physicochemical and bioactive properties of the CA/shikonin nanofiber mats were evaluated. Scanning electron microscopy (SEM) was used to characterize the surface morphology of the CA and CA/shikonin nanofibrous mats and their average diameters ranged from 1300 to 1330 nm. The CA/shikonin nanofiber mats exhibited suitable equilibrium water content (EWC) and water vapor transmission rate (WVTR). The antioxidant activity was evaluated with the DPPH assay. The bacterial activity of the CA/shikonin nanofiber mats against Escherichia coli and Staphylococcus aureus was evaluated by the zone of inhibition method and the result showed good antibacterial activity. In vitro indirect MTT assay also showed that the CA/shikonin nanofiber mats have good cytocompatibility to L929 mouse fibroblasts. The results indicate that CA/shikonin nanofiber mats could be potentially used as novel antibacterial wound dressings.
