Arecoline和CuCI2對中國倉鼠卵(CHO)細胞之細胞:遺傳毒性之研究

Mutual Interaction of Arecoline and CuCl2-Induced Cytogenotoxicity on Chinese Hamster Ovarian Cells

摘要


本研究利用微小核(MIN)法來探討檳榔塊成分(CuCl2和arecoline)在中國倉鼠卵(CHO)細胞之遺傳毒性。並分別以MTT法和核分裂指數(nuclear division index: NDI) 探討arecoline和CuCl2在CHO細胞之細胞毒性。此外,以流體細胞測定法(flow cytometry)探討arecoline和CuCl2影響CHO細胞之細胞凋亡及細胞週期之分佈情形。 遺傳毒性的研究結果顯示arecoline及CuCl2均如adriamycin有統計意義的增加CHO細胞之微小核(MN)數。交互遺傳毒性之探討發現CuCl2可加強arecoline之遺傳毒性。MTT之研究結果發現高濃度之arecoline和CuCl2均能抑制CHO細胞之存活率,但CuCl2不加強arecoline之細胞毒性。此外,arecoline和CuCl2對CHO細胞之NDI的影響較複雜:arecoline在低濃度加強NDI,高濃度則呈現抑制作用;CuCl2則增加NDI。細胞凋亡之研究結果顯示,arecoline和CuCl2均不造成細胞凋亡,亦不影響細胞週期各期的分佈、細胞體積的細胞之顆粒度。結論是arecoline和CuCl2在CHO細胞均呈現細胞毒性且非以細胞凋亡方式殺死CHO細胞。二者在CHO細胞亦具有遺傳毒性,CuCl2且可增強arecoline之遺傳毒性,可能因此促進arecoline之口腔癌病變作用。

In this study, the micronucleus (MN) test, MTT assay, nuclear division index (NDI) and flow cytometry were conducted to assay genotoxic, cytotoxic and apoptotic effects of arecoline and CuCl2 on Chinese hamster ovarian (CHO) cells. Results of the MN test indicated that both arecoline and CuCl2 significantly increased the number of micronucleated cells. CuCl2 potentiated the genotoxicity of arecoline. MTT assay showed that both CuCl2 and arecoline at high concentrations significantly decreased the viability of CHO cells; however, CuCl2 did not affect the cytotoxicity of arecoline. The NDI results were complex: at a lower concentration of 0.03 μM, arecoline increased nuclear division; however, at a higher concentration of 10 μM, arecoline depressed nuclear division. Regardless, 100 μM CuCl2 increased NDI. Flow cytometry demonstrated that neither Cu2+ nor arecoline produced hypoploid cells or changed cell cycle distribution, cellular size or cellular granularity. Obviously, both Cu2+ and arecoline did not cause apoptosis in CHO cells. Based on these findings, it is concluded that both CuCl2 and arecoline were genotoxic and cytotoxic to CHO cells, and that CuCl2 increased arecoline induced-genotoxicity. Thus, CuCl2 seems to contribute to the oral mutagenic effects of arecoline.
