紫外線在我們生活環境中無所不在,適量的吸收紫外線有助於維他命D的合成,而暴露過量則會造成皮膚曬傷、自由基產生、發炎及老化,嚴重甚至致癌。因此本研究除了要探討綠球藻萃取液的抗氧化作用外,也要研究綠球藻萃取液對抗紫外線傷害的作用。實驗結果顯示,綠球藻萃取液具有良好的抗氧化作用包括清除超氧陰離子、還原力及螯合亞鐵離子的能力,其中又以清除超氧陰離子的效果最佳EC_(50) 0.217%。此外,綠球藻萃取液可以促進纖維母細胞生長達24%。在UVB誘發的光毒性實驗中,0.05%, 0.5%及1%的綠球藻萃取液對於UVB 30, 60及120 mJ/cm^2引發的細胞毒性有顯著的保護作用。細胞傷口癒合試驗也顯示經0.5%綠球藻萃取液處理的細胞在24小時後,有促進細胞癒合,其癒合速度比控制組快了39.43%。總言之,綠球藻萃取液具有很好的抗氧化、促進細胞生長並免於紫外光傷害的功效,可開發為對抗光毒性或老化的化妝品原料。
UV is omnipresent in our environment. Absorption of ultraviolet (UV) light will help vitamin D synthesis. However, exposure to excessive UV radiation can lead to sunburn, free radicals production, inflammation and aging, even cancer. Therefore, the aim of this study is to explore the antioxidation effects of chlorella extract, and the role on against UV radiation induced damage. The results showed that the chlorella extract had very good antioxidation effect, including the ability to remove superoxide anion, reducing power and chelating ferrous ions, and the best of them was the effect of removing superoxide anion EC_(50) 0.217%. In addition, chlorella extract can promote fibroblast growth of 24%. In the UVB-induced phototoxicity test, 0.05%, 0.5% and 1% chlorella extract had a significant protective effect on UVB-induced phototoxicity at 30, 60 and 120 mJ/cm^2, respectively. The cell wound healing assay also showed that cells treated with 0.5% chlorella extract had a percent closure of 39.43% faster than the control group after 24 hours treatment. In general, the chlorella extract has good antioxidation effects; can promote cell growth and prevent cells from UV radiation induced-cell damage. Chlorella extract can be developed as a cosmetics raw material against UV-induced phototoxicity or photoaging.
