水溶性幾丁聚醣作為組織抗沾黏複合材料之製備與評估

Preparation and Evaluation of PEG-Chitosan Hybrid as Tissue Anti-Adhesion Materials

摘要


於本實驗經由一端醛基化的甲氧基聚乙二醇(MPEG)與幾丁聚醣(Chitosan)的胺基進行接枝化合反應,製備出聚乙二醇與幾丁聚醣的複合共聚物(PEG-Chitosan hybrid),以傅利葉紅外線光譜儀(FT-IR)、核磁共振儀(H-NMR)、熱分析儀(DSC)、接觸角測試及細胞流動室等測試進行高分子共聚材料的定性分析與體外生物相容性評估。而藉由分子量、接枝率的不同條件得到各種相對的材料特性,將其製備成薄膜的形式,應用於組織抗沾黏與降低細胞吸附。由實驗結果顯示H2-75,H2-50,L2-50三種條件下所製備出的薄膜其成膜性最佳,且對於親水性與抗細胞吸附能力皆有良好的效果。因此在未來我們將藉由動物體內實驗加以印證此三種條件的共聚複合膜對於組織抗沾黏方面有良好的功效。

In this study, we modify different molecular weight of PEG by oxidizing the hydroxyl group into aldehyde, and then react with the amine groups of chitosan. These prepared PEG-chitosan hybrids were then cast into films and characterized by FTIR, DSC and contact angle measurements. The preliminary results showed that this modified hybrid has lower contact angle as compared with the untreated chitosan film (decreased from 75° to 18°~60°). In cell adhesion test, cells less adhered to the hybrid film than glass and chitosan film.
