本研究主要探討錦葵科植物台灣山芙蓉及豆科植物菲律賓千斤拔兩種植物根部之化學成分,以了解植物中的成分與降血糖活性之間的關連性。將兩個植物萃取物溶於水,且再以n-hexane、EtOAc或n-butanol等不溶於水的溶媒,分別從較低極性到較高極性進行分配萃取,各劃分層再經過多種管柱層析及不同溶媒系統分離純化後,總共分離純化出三十一個化合物,並將所分離得到的化合物進行降血糖活性測試。從台灣山芙蓉根部之70% acetone / H2O之萃取物之正己烷層得到isohemigossypol-1-methyl ether (9) ; 由乙酸乙酯層分離出protocatechuic acid (1)、p-coumaric acid (2)、caffeic acid (3)、ferulic acid (4) ; 從正丁醇層分離出uracil (5)、uridine (6)、adenosine (7)、guanosine (8)、rutin (10)。另外,菲律賓千斤拔根部75% EtOH / H2O之萃取物,於乙酸乙酯層中分離純化出的化合物為genistein (11)、prunetin (12)、2'-hydroxygenistein (13)、genistin (14)、ononin (15)、dalbergioidin (16)、naringenin (17)、flemichin D (18)、khonklonginol H (19)、dorsmanin I (20)、aromadendrin 6-C-glucoside (21)、4'-O-methylgallocatechin (22)、5, 7, 2', 4'-tetrahydroxy-3-arylcoumarin (24)、aureol (25)、gallic acid (31);由正己烷層分離出eriosematin (23)、??-sitosterol (26)、stigmasterol (27)、??-sitosterol glucoside (28)、stigmasterol glucoside (29)、lupeol (30)。生物活性部分在C2C12細胞株測定葡萄糖攝入量的活性試驗中,發現2、3、4、7、24、30具有促進作用。
In this study, antihyperglycemia bioactive constituents of Hibiscus taiwanensis Hu and Flemingia philippinensis Merr. et Rolfe were investigated. Extract that dissolve in water partitioned with solvent which was insoluble in water from low polarity to high polarity gradually. Through all kinds of different solvents and chromatography system, total of compounds were isolated. Finally, glucose uptake of antihyperglycemia activity were tested by compounds which are isolated from those plant. Total of 31 compounds were isolated. Ten compounds which have been isolated from n-hexane layer, ethyl acetate, and n-butanol layer of 70% acetone / H2O extract of Hibiscus taiwanensis Hu are protocatechuic acid (1)、p-coumaric acid (2)、caffeic acid (3)、ferulic acid (4)、uracil (5)、uridine (6)、adenosine (7)、guanosine (8)、isohemigossypol-1-methyl ether (9)、rutin (10). Twenty-one compounds which have been isolated from ethyl acetate and hexane layer of 75% EtOH / H2O extract of Flemingia philippinensis Merr. et Rolfe are genistein (11)、prunetin (12)、2'-hydroxygenistein (13)、genistin (14)、ononin (15)、dalbergioidin (16)、naringenin (17)、flemichin D (18)、khonklonginol H (19)、dorsmanin I (20)、aromadendrin 6-C-glucoside (21)、4'-O-methylgallocatechin (22)、eriosematin (23)、5, 7, 2', 4'-tetra-hydroxyl-3-arylcoumarin (24) 、aureol (25)、??-sitosterol (26)、stigmasterol (27)、??-sitosterol glucoside (28)、stigmasterol glucoside (29)、lupeol (30)、gallic acid (31). Compounds 2、3、4、7、24、30 can enhance glucose uptake in C2C12 cell line.
