Effect of Photosynthetic Photon Flux Density and Sucrose on the Photoautotrophy of Rose Explants in Vitro

摘要

The growth of the explants of Rosa hybrida cvs. 'Samantha' and 'Landora' shoots cultured under higher (700 μmol m^(-2) sec^(-1)) photosynthetic photon flux density (PPPD) was better than those cultured under lower (35 μmol m^(-2) sec^(-1)) PPFD. Under high PPPD, explants collared on medium, containing 15g/l sucrose, got the highest fresh weight. About 40% of 'Samantha' explants cultured under high and medium, without sucrose, survived at the end of the 4(superscript th) week. However, under the same condition only 50% of 'Landora' explants survived at the end of the 3(superscript th) week. After the shoot explants of 'Samantha' and 'Landora' were cultured under high PPFD and medium, containing 15g/l sucrose for 4wks, the stable concentration of CO2 in culture vessel reached 0 ppm during light period. However, under tow PPFD, the stable concentration of CO2 in vessel, which 'Samantha' or 'Landora' explants were cultured, was 130 or 420 ppm, respectively. When the sucrose in the medium was increased to 30g/t, 'Samantha' or 'Landora' explants were cultured under high PPFD the CO2 concentration in vessel was high to 620 or 830 ppm. When the explants of 'Samantha' cultured on medium without sucrose nod under high PPFD the CO2 concentration difference between light and dark periods increased as well as the days of culture extended. Furthermore, after a four-week culture, concentration during light time was as be as l00 ppm only. However, 'Landora' explants cultured on medium without sucrose, no differences were observed in the CO2 concentration between light and dark periods, whitchever explants were under high or low PPFD.
