Effects of N^6-benzyladenine, Organic Additives, and Sucrose on Shoot Growth of Cymbidium ensifolium

摘要


本研究探討培養基成分之細胞分裂素BA、有機添加物及蔗糖對四季蘭‘金荷’芽體生長之影響。結果顯示四季蘭‘金荷’之芽體培養於含BA之1/2MS培養基,可促進芽體分化與生長,避免芽體逆生長為根莖組織,使其生育整齊,BA處理濃度以3 mg.L^(-1)最為適宜。有機添加物以馬鈴薯泥搭配香蕉泥之組合對促進芽體生長之效果較單一有機添加物香蕉泥或馬鈴薯泥處理為佳,其中以80 g.L^(-1)馬鈴薯泥搭配40 g.L^(-1)香蕉泥處裡之芽體抽長數最多。培植體以同一根莖帶3芽處理之芽體抽長數高於帶5芽處理者。蔗糖濃度以20或40 g.L^(-1)較為適宜,高濃度(80 g.L^(-1))之蔗糖顯著促進根系生成與生長,不利於日後瓶苗出瓶作業。根莖誘導芽體分化階段,以前處理0.5或1 mg.L^(-1) BA其芽體生育速度較0.1-0.5 mg.L^(-1) TDZ處理組快速,且芽體生長整齊,無褐化情形發生。

This study investigated the effects of BA, organic additives, and sucrose in 1/2 MS basal medium on bud growth of Cymbidium ensifolium (L.) Swartz 'Jin-he'. Results show that BA could promote the differentiation and growth of rhizome-derived shoots or shoot buds, and prevent the shoot buds from turning to rhizomatous tissue. BA concentration at 3 mg.L^(-1) resulted in more uniform shoots. Combinations of organic additives of potato and banana homogenate had more shoots than single organic additives, with highest elongated shoots in 80 g.L^(-1) potato combined with 40 g.L^(-1) banana homogenate. The rhizome explant with three buds had more elongated shoots than rhizome with five buds. Sucrose concentration at 20 or 40 g.L^(-1) were optimum for shoot growth, while 80 g.L^(-1) sucrose treatment stimulated root formation that caused inconvenience for de-flask. Shoots or shoot buds were uniform, no browning, and grew faster in medium pretreated with 0.5 or 1 mg.L^(-1) BA, as compared with 0.1-0.5 mg.L^(-1) TDZ.
