短暫浸漬系統在麗格秋海棠(Begonia×hiemalis)大量繁殖之研究

Mass Propagation of Riger Begonia (Begonia×hiemalis) by Temporary Immersion Systems

摘要


本試驗主要建立麗格秋海棠大量繁殖之組織培養方法,探討短暫浸漬系統(Temporary immersion system)對麗格秋海棠葉片誘導之芽體增殖與植株再生之影響。以葉片為培殖體接種於含有5.4μMNAA及2.22μMBA的MS培養基,培養六週後,將叢生之不定芽連同培殖體,繼代培養於短暫浸漬系統,探討浸漬液體體積與不同浸漬頻度(hr/cycle)對芽體增殖與植株再生之影響。試驗結果顯示,每個培養容器注入200mLMS液體培養基,每6小時進行浸漬30秒為適當之條件,每葉片之培殖體可產生33.3株再生植株,其中62%的再生植株其株高分布於2.0~2.9cm,平均2.1cm,較傳統的固體培養或液體培養,可獲得較均質的植株。因此,以本研究建立之麗格秋海棠短暫浸漬培養系統,可做為未來大量繁殖商業生產之基礎。

The purpose of this study was to establish a protocol for mass propagation of (Begonia×hiemalis) and to investigate the effects of the temporary immersion system (TIS) on adventitious shoot growth and plant regeneration. The adventitious shoots were induced from leaf explants cultured on MS medium supplemented with 5.4 μM NAA and 2.22 μM BA for 6 weeks and then subcultured with the temporary immersion system. Multiplication rate and plant regeneration of adventitious shoots were examined within different immersion frequencies (hr/cycle) and the volumes of TIS per vessel. The optimum parameters of the temporary immersion system were 200 mL liquid medium per vessel and 30 sec immersion per 6 h. The average plantlet number per leaf explants was 33.3, which was higher than traditional liquid and solid cultures. The height of 62 % plant ranged from 2 cm to 2.9 cm. TIS provided a more efficient mass proliferation of Riger begonia.
