乙烯及離層酸抑制劑對大豆雜交胚及莢果柄離層組織發育之影響

Effects of Ethylene and Abscisic Acid Inhibitors on the Hybrid Embryos from Interspecific Crossing between Glycine spp.

摘要


為探討大豆種間雜交莢果當施用乙烯及ABA抑制劑處理後,對雜交胚發育及雜交莢果柄分裂層形成之影響。本試驗以臺灣之野生種大豆,臺東野生大豆(Glycine dolichocorpa)、闊葉大豆(G. tomentella)及澎湖大豆(G. tabacina)與栽培種大豆高雄選10號(G. max cv. KS10)進行大豆種間雜交,並於雜交授粉後噴施促進花粉管伸長之GAs+NAA+Kinetin,不同濃度之乙烯抑制劑AgNO3、CoCl2及ABA抑制劑aminotriazol (ATZ),並以ethrel、ABA及對照進行比較,藉以觀察雜交胚及莢果柄離層組織發育,其結果如下:施用乙烯及ABA抑制劑後,對莢果柄及雜交胚發育,經由石蠟切片觀察,AgNO3 100mg L-1及CoCl213mg L-1均可促G. max x G. dolichocarpa至授粉後33天左右莢果柄分裂層才有異常情形,相較對照於27天左右掉落,明顯有延遲落莢情形。而G. max x G. tomentella及G. max x G. tabacina之雜交組合亦有相同結果。ABA 合成抑制劑ATZ 施用於G. max x G. dolichocarpa之雜交組合,莢果柄在15-21天才開始產生分裂層,而對照之莢果柄分裂層已快形成,顯示ABA合成抑制劑確可抑制脫落延遲落莢,但與乙烯抑制劑比較,ABA合成抑制劑不如乙烯作用、合成抑制劑抑制分裂層形成之效果明顯。

The objectives of this study were to determine the effects of ethylene and abscisic acid (ABA) inhibitors on the development of hybrid embryos and the formation of abscission layers of the hybrid pod peduncles from interspecific crossing between Glycine spp.. The hybridization crosses between Glycine max cv. KS10 and the native species, i.e., G. dolichocarpa, G. tomentella and G. tabacina, were used in this study. For comparing the development of hybrid embryos and the formation of abscission layers of podpeduncles, the solution of GAs+NAA+Kinetin, ethylene inhibitors (i.e., AgNO3 and CoCl2), and ABA inhibitor [aminotriazol (ATZ)] were applied after pollination as compared with those applied with ethrel, ABA and control treatments. The pod peduncles and hybrid embryos after treating with ethylene and ABA inhibitors showed that the abscission layer of G. max x G. dolichocarpa formed at 33 days after pollination (DAP), while the control formed at 27 DAP. The results showed that the inhibitors could delay the hybrid pods of G. max x G. dolichocarpa to fall down. The results of G. max x G. tomentella and G. max x G. tabacina were the same as that of G. max x G. dolichocarpa. The abscission layers of hybrid pod peduncles of G. max x G. dolichocarpa treated by ATZ started to form at 15-21 DAP, while those of the control had already formed. The results showed that the effect of inhibitor of ABA synthesis to delay formation of abscission was less than that of inhibitors of ethylene action and synthesis.
