多光子雷射掃瞄顯微系統之架設與脂肪肝診斷

摘要


肝相關疾病在台灣是極為重要的疾病。早期預防的機制仍存在許多發展空間。在這個研究中我們首先利用非線性光學顯微技術觀察在肝病變發展初期特徵之一的脂肪肝現象。透過對肝臟膠原蛋白增生的影像分析,量化脂肪肝導致纖維化的行為。 非線性光學顯微術由於它的非侵入性和光學切片能力成為觀測生物樣品極為有用的技術。除此之外,我們可以自由的透過分光元件的搭配,取得特定波段的的螢光及倍頻以突顯特定部位的訊號。因此在有了量化膠原蛋白增生的初步結果後,我們利用光參數震盪雷射提供的近紅外光源(1.1~1.35μm)嘗試搭建一套新的雷射掃描顯微系統,期望在掃描中同時取得生物組織自發螢光、二倍頻及三倍頻訊號。有助於我們更加了解疾病的演化過程。

Liver related diseases are major illnesses in Taiwan. There are many chances for us to improve the examination of them in early stages. Fatty liver disease is one of the usual courses during these illnesses, so in this thesis we first used nonlinear optical microscopy to research normal and fatty liver tissues in this investigation. From imaging and analyzing the collagen overgrowth in liver, we can quantify the fibrosis due to fatty liver disease. Nonlinear optical microscopy is a valuable technique for observing biological samples due to its non-invasive nature and optical sectioning ability. Besides this, we can get specific fluorescence and harmonic generation signals arbitrarily due to dichroic units by our own choices. Accordingly, we try to set up a new laser scanning microscopic system with near infrared light source (wavelength :1.1~1.35μm ) from the optical parametric oscillator. We wish to acquire autofluorescence, second harmonic generation, and third harmonic generation signal from biological samples simultaneously for helping us understand the dynamics of diseases.
