金合歡醇應用於皮膚修復材料之評估

Evaluation of using farnesol in skin repairs: in vitro and in vivo studies

摘要


金合歡醇具有刺激纖維母細胞增生膠原蛋白之特性,目前多數用於妝品類產品做添加香氣應用,尚未有研究探討其應用於曬傷及燒燙傷傷口修復之效用。本論文研究動機與目的為評估金合歡醇之膠原蛋白增生能力及含金合歡醇複合凝膠於皮膚傷口修復之效用。實驗中評估金合歡醇其濃度於0.1~0.3 mM不具細胞毒性,且能刺激纖維母細胞膠原蛋白脂增生;利用羥丙基甲基纖維、透明質酸及三仙膠製備複合凝膠之傷口敷料,結果顯示複合凝膠能阻擋大部分UVB及避免纖維母細胞受到刺激發生週期改變;曬傷動物模型之皮膚修復結果顯示其金合歡醇複合凝膠有助於降低傷口炎症反應及膠原蛋白增生。為能提高金合歡醇之藥物濃度及藥物緩釋時間,本論文以微脂粒來包覆金合歡醇應用於燒燙傷傷口治療。實驗結果顯示包覆後金合歡醇可達48小時緩釋時間,且燒燙傷動物實驗結果以微脂粒化金合歡醇複合凝膠顯示其能降低傷口之發炎因子IL-6表達及促進膠原蛋白之增生。

Farnesol is, a natural 15-carbon organic compound, has been found to increase the synthesis of collagen of fibroblasts. It is often used as an ingredient in cosmetic products, less in the applications of wound healing from sunburn and moderate/heavy burns. In the current study, the capability of farensol in collagen synthesis and farensol-containing gels were designed to evaluate the repairing effects on injured skin. The obtained results the farensol is non-toxic under the concentration of 0.3 mM and able to stimulate fibroblasts to secret collagen. Gel containing hydroxypropyl methylcellulose (HPMC), hyaluronan (HA), xanthan gum (XG) and farnesol revealed that could occlude the UVB light and retain the cell cycle of fibroblasts. The sunburn animal study results also indicated that the prepared gels could decrease the inflammatory effect after sunburn and increase the collagen synthesis. A plural gel containing farensol liposomes (using as a slow release agent) was also prepared to evaluate the healing effects of farensol liposomes in burn-injured rats. The results indicated that the liposomal farensol has non-toxic characteristics, the farensol liposomes exhibited good spherical shape and with average size of 100 ± 30 nm in diameter. The 31% of encapsulated farensol released after 48 hours incubation. Importantly, the gel containing liposomal farensol decreased the IL-6 factor expression and stimulated the fibroblasts synthase the collagen after burn injury in SD rats.
