飲料中蘆薈素檢驗方法之探討

Determination of Aloin in Drinks by HPLC

摘要


本計劃針對飲料中的廬薈素,採用HPLC分析方法加以檢測。HPLC的分析條件使用CH3OH/H2O(40:60, V/V)為移動相,以1ml/min的流速,流經Lichrosorb RP-8(5μm)的管柱(150×4mm)並於波長295nm下測得樣品層析圖,再和標準品所得結果比對。本方法之最低檢測量為0.04ppm。外添加廬薈素的回收實驗結果,以汽水為例:0.05-0.20μm/ml範圍內三種濃度之回收率為86.81-97.92%,變異係收為2.95-5.91%,其標準曲線及回收曲線之相關係數分別為1.0000和0.9972。另以運動飲料為例,其0.05-0.20μg/ml範圍內之外添加回收率為110.73-116.70%,變異係為3.27-6.48%,其標準曲線及回收曲線之相關係數分別為0.9997和0.9994。故本檢驗方法於風味皆由添加香料所提供之清涼飲料類如汽水或運動飲料等,其中若有超出規定之微量蘆薈素,皆可檢測出來。目前,行政院衛生署規定“飲料中廬薈素的含量為0.1ppm以下”。由市面上探樣50件清涼飲料,以上述檢驗方法均未檢出蘆薈素。

A high perfermance Liquid chromatographic method was described for determination of aloin in drinks at this report. Aloin was extracted and cleaned up through disposable cartridage (Adsorbex RP-18) using methanol-water (55/45, V/V) as eluent. The eluted aloin was separated and detected by HPLC on Licrosorb RP-8 column with methanol-water (40/60, V/V) mobile phase, and measured at 295nm. The Limit detection level of aloin by this method was 0.04ppm. When aloin added to sport drinks and soft drinks at the levels of 0.05-0.02g/ml, the recovery were 86.81-97.92% from the former and 110.73-11630% from the latter. the survey of aloin by this HPLC method resulted that aloin contents were not violated the regulation in 50 marketed drinks which collected from supermarkets in Taipei from Nov. 1988 to May 1989. The legal allowance amount for aloin in drinks was set at 0.1mg/kg by the Health Department.
