小分子化合物庫之建構邏輯及合成方法

The Synthetic Approach and Logic of Design Behind the Small Molecule Compound Library Synthesis

摘要


目前在化學生物學領域中,經常利用小分子化合物來對生物系統進行研究,此研究稱為化學遺傳學。研究中所發現具有生物活性的分子,或者未知作用的蛋白質,往往可以提供藥物化學做更深入的研究。在此當中,小分子化合物庫的品質好壞,將左右研究的成敗與否。此篇文獻回顧,將近期廣泛被使用的化合物庫構建方法進行統整,依照分子庫建立的邏輯及方法分類,並且挑選具有代表性的例子做為參考。希望能夠提供科學研究人員,對於這個領域通盤性的瞭解,進一步的應用現有的方法,或者開發出更為新穎有效的化合物庫合成邏輯方法。

In the field of chemical biology, small molecules have been widely utilized for the elucidation of biological systems, named chemical genetic approaches. The chemical genetic approaches are allowed to uncover the bioactive small molecules and function-unknown proteins that can serve as the starting point for further medicinal chemistry investigations. Therefore, the quality of small molecule compound library determines the fate of biological research outcomes. In this article, we summarized advanced and frequently used approaches for the compound library synthesis. The classification of these approaches based on the logic of preparation. Besides, we also selected representative examples for each approach for the better understanding. Hopefully, this review article can provide a general view for the field, the ideal of application these approaches or the inspiration of developing novel small molecule library synthetic approach.
