用於“識別”及“治療”癌症腫瘤細胞的放光過渡金屬錯合物:設計及應用探討

Phosphorescent Transition Metal Complexes for Cancer Theranostics: Design, Challenges and Opportunities

摘要


磷光過渡金屬錯合物(phosphorescent transition metal complexes, PTMCs)過往被廣泛用於顯示器中,但應用在生醫材料領域就少得多;隨著檢驗技術進步,將分子診斷技術做為「識別」腫瘤細胞與「治療」串接達到精準醫療的概念發展越來越快速。PTMCs具有易於修飾的立體結構、較長的生命週期、放光可拓及近紅外光區、光穩定性佳等優點,若將顯影成像、治療及藥物輸送等功能結合在一起,在材料發展上將是一個新的突破。本文介紹以PTMCs分子識別腫瘤細胞的方法,包含酵素裂解法、缺氧細胞的腫瘤顯影成像法,以作為識別腫瘤細胞癌症的探針;接著針對PTMCs分別運用在化學治療法、光動力療法、標靶治療法及綜合治療法中,對腫瘤細胞治療做一些整理介紹。最後再針對分子的設計做出一些建議。

Phosphorescent transition metal complexes (PTMCs) have been most widely used in OLEDs, but few in biomaterials. Theranostics approach is a combination of specific targeted therapy based on specific targeted diagnostic tests. Theranostic agents can be designed to personalize treatment toward precision medicine of cancer. PTMCs have attracted recent attention due to their advantages of large Stokes shifts, long lifetimes, visibe or near-infrared emission, high photostability and ease of structural modifications. Representative examples of design strategies, challenges and opportunities for PTMCs for cancer diagnostics and theranostics are discussed in this articles.
