Ultrastructure of the Aecial and Pycnial Stages of Pllccillia dioicae, the Cause of Leaf Rust on Lettuce

摘要


利用掃描及穿透式電子顯微鏡檢視萵苣葉銹病之春胞子及精胞子之微細構造。精子器內具有短柵狀排列之精子器柄,其上著生表面平滑及橢圓形之精胞子。精子器柄尖端具有多層之環狀結構。春胞子器之被膜(peridium)呈杯狀,由單層細胞所組成。被膜之單一細胞呈長菱形,其表面有棍棒狀之突起。春胞子淡黃色,近乎球形,除了其上圓滑地(tonsure)外,表面著生了小瘤狀之突起。春胞子具有4至5個發芽孔,其上被“栓塞物”(plug)所覆蓋著,發芽孔環列於春胞子之中腰部份。間隔細胞位於鏈狀排列之春胞子間,其表面無突起,在春胞子釋放前被解體。本文係萵苣菜直銹病微細構造之首次報導。

The surface markings of the spores from the pycnial and aecial stages of Puccinia dioicae Magn. on leaves of lettuce (Lactuca sativa L.) cv. Buttercrunch were examined using scanning and transmission electron microscopy. The pycnium had a short palisade of pycniosporophores, which produced smooth, ellipsoidal pycniospores. Multilayered collars were observed at the apex of the pycniosporophores. The large, cupulate aecium was borne within a well-developed peridium, which was one cell-layer thick. The peridial cells were rhomboidal, with club-shaped ornaments. The pale aeciospores were nearly globoid and densely ornamented, with smooth. knoblike verrucae, except in the smooth circular patch (tonsure) areas. Four to five germ pores covered with large, well-formed plugs were equatorially arranged on the aeciospores. The intercalary cells, which resided between each of the aeciospores in the chain, were thin-walled, free of ornamentation, and disintegrated prior to aeciospores discharge. This is the first report of the detailed ultrastructural morphology of this fungus.
