台灣綠竹種內遺傳相似性

Genetic Similarity Detected by Molecular Markers in Green Bambooes (Bambusa Oldhamii Munro) of Taiwan

摘要


本研究以逢機擴增多型性DNA(RAPD)、切割擴增多型性序列(CAPS)與簡單重複序列間產物(ISSR)三種分子標誌分析栽培綠竹(Bambusa oldhamii)之遺傳相似性。綠竹樣本包括由桃園區農業改良場在北部不同縣市綠竹園收集之綠竹、台南區農業改良場所保留之無病毒綠竹、以及自嘉義市、台南關廟、宜蘭與屏東海豐取樣之綠竹。72個參試材料中,除了一個混雜在桃園區農業改良場台北分場之烏腳綠竹(B. edulis)樣本具有多型性差異外,71個綠竹樣本之三種分子標誌都只表現單型性。來自不同產地之綠竹樣品沒有遺傳差異;不同群體之綠竹並未檢視出多型性。

Random amplified polymorphic DNA (RAPD), cleaved amplified polymorphic sequence (CPAS) and inter simple sequence repeat markers (ISSR) assays were applied to detect the molecular variation among green bamboo (Bambusa oldhamii) accessions under cultivation in various regions. A total of 72 accessions were collected from Taoyuan District Agricultural Research and Extension Station (originally collected from different townships in northern Taiwan), Tainan District Agricultural Research and Extension Station, and cities of Chiayi, Tainan, Yilan and Pingtong. Only monomorphic fragments were generated by all three molecular markers in 71 B. oldhamii accessions and one accession of B. edulis was detected and identified by its polymorphism variation. There was no intra-specific polymorphism among B. oldhamii accessions from different populations in Taiwan.
