紅鳳菜及紅莧菜之抗氧化、抗突變及調節小鼠脾臓細胞分泌性能研究

Antioxidant, Antimutagenic and Immunomodulatory Potentials of Gynura Bicolar and Amaranthus Gangetcaue

摘要


本研究選取臺灣產紅色蔬菜紅鳳菜與紅莧菜,探討其對亞麻油酸之抗氧化性、對致突變物IQ之抗突變性及對小鼠初代脾臓細胞之名疫調節功能,結果發現選取之二種紅色蔬菜水萃取物均具強抗氧化性,紅莧菜之抗氧化性優於紅鳳菜。不論是否加熱,其抗氧化力均較同濃度維生素E佳。在抗突變性方面,紅鳳菜及紅莧菜水萃取物之抗突變能力則遠不及同濃度之BHA及維生素E。紅鳳菜及紅莧菜榨汁液與BALB/c雌鼠脾臓免疫細胞共同培養,結果發現僅紅鳳菜榨汁具有刺激脾臓初代細胞增生能力,但經加熱及活性碳脫色後則活性消失。以酵素免疫分析法(ELISA)分析脾臓細胞分泌Th1(IFN-γ及IL-2)及Th2(IL-4、IL-5及IL-10)細胞激素之變化,結果發現紅鳳菜及紅莧菜榨汁原液具有調節小鼠脾臓Th1及Th2細胞激素分泌之能力,添加紅鳳菜及紅莧菜榨汁使小鼠脾臓細胞促發炎細胞激素TNF-α之自發性分泌量降低,脾臓細胞在經脂多醣(LPS)刺激後,添加紅鳳菜粗榨汁液可降低發炎媒介物前列腺素E2(PGE2)之分泌,紅莧菜則無法逆轉LPS所誘發之PGE2生成。綜合評估本試驗所選取之二種紅色蔬菜之生理調節功能,紅鳳菜及紅莧菜均具有抗亞麻酸氧化作用,抑制小鼠初代脾臓細胞TNF-α之自發性分泌,顯示兩者均具有抗發炎作用,但整體評估則以紅鳳菜較具生理調節功能。

To determine whether two species of red-colored vegetables, Gynura bicolar and Amaranthus gangetcaue, have the antioxidant, antimutagenic, and immunomodulatory potentials, we measured the linoleic acid oxidation level, Salmonella typhimurium mutagenicity of 2-amino-3methylimidazo[4,5-ƒ]quinoline(IQ) and changes of primary splenocyte culture from female BALB/c mice by Gynura bicolar and Amaranthus gangetcaue. We found that water extracts of the two species of the two species of red-colored vegetables we chosen had strong antioxidant activity. Both of Gynura bicolar and Amaranthus gangetcaue were superior to α-tocopherol at the same concentration for the 72-h duration of the linoleic acid oxidation experiment. Amaranthus gangetcaue whether heat-treated or not had the better antioxidant activity than that of Gynura bicolar. However, both of Gynura bicolar and Amaranthus gangetcaue showed much lower antimutagenicity as compared with that of the same concentration of BHA and α-tocopherol. Only the vegetable juice of Gynura bicolar could stimulate spleen cells to proliferate, although the activity disappeared after it was treated with heat or activated charcoal. The secretions of Th1- (IFN-Υand IL-2) and Th2-type (IL-4, IL-5 and IL-10) cytokines from primary splenocyte culture were assayed by ELISA. It was found that both vegetable juices of Gynura bicolar and Amaranthus gangetcaue modified the secretion of Th1 and Th2 cytokines of primary splenocyte culture, however the spontaneous secretion of TNF-α, a pro-inflammatory cytokine, decreased. The vegetable juice of Gynura bicolar inhibited the inflammatory mediator, prostaglandin E2(PGE2),production of primary splenocyte culture stimulated by lipopolysaccharide (LPS), but Amaranthus gangetcue could net reverse the inflammation status . These results suggest that both of Gynura bicolar and Amaranthus gangelcue have antioxidant and anti-inflammatory activity, however Gynura bicolar has a better physiologic function to us.
