運用高效液相層析法同時檢測飼料中安默西林與安比西林

Simultaneous Determination of Amoxicillin and Ampicillin in Animal Feed by High Performance Liquid Chromatography

摘要


本實驗的目的在建立高效法相層析法同時檢測飼料中安默西林(amoxicillin)及安比西林(ampicllin)的方法。層析管柱使用SymmetryShield(superscript ®) column(RP8,5 μm,I.D. 4.6 × 250 mm)。移動相為0.01 M磷酸二氫鉀緩衝液(pH 2.5):乙腈(85:15,v/v),螢光檢測器檢測波長設定為λem 358 nm與λem 440 nm。安默西林及安比西林均以三種不同濃度添加於飼料中,平均回收率介於102.16 %及113.60 % 間;回收率之測試日內及測試日間變異係數皆在3.10 %以下。安默西林及安比西林之檢測極限分別為0.073及0.003 mg/kg,定量極限分別為0.242及0.009 mg/kg。 本試驗所建立之檢測方法通過準確度、精密度、專一性等各項確效試驗,可實際應用於檢測飼料中安默西林及安比西林之含量。

The purpose of this study was to develop an analytical method using high performance liquid chromatography (HPLC) for simultaneous determination of amoxicillin and ampicillin in animal feed. A SymmetryShield(superscript ®) column (RP8, 5 μm, 4.6 X 250 mm I.D.) was selected and a mobile phase consisting of 0.01 M potassium dihydrogen phosphate (pH 2.5): acetonitrile = 85:15 (v/v) was used. Fluorescence detector was set at excitation wavelength of 358 nm and emission wavelength of 440 nm. The average recovery ranges of amoxicillin and ampicillin from three different spiked concentrations in animal feed were between 102.16 and 113.60 %. The intra-day and inter-day repeatability were below 3.10 % relative standard deviation. for both compounds. The limits of detection of amoxicillin and ampicillin were 0.073 and 0.003 mg/kg and the limits of quantification were 0.242 and 0.009 mg/kg for fortified animal feed, respectively. The specificity, precision, accuracy, and reproducibility were also performed and confirmed in this analysis method. This assay can be routinely used to detect amoxicillin and ampicillin in animal feeds.
