The Anesthetic Effects of Different Clove Essence Concentration Combined with Two Temperature on Persian Sturgeon, Acipenser persicus

不同濃度的丁香精油於兩種溫度下對波斯鱘Acipenser persicus的麻醉效果

摘要


本實驗探討丁香精油作為麻醉劑對波斯鱘Acipenser persicus的麻醉效果,以及丁香精油對血漿中的腎上腺皮質醇與葡萄糖濃度、血液學因子(血紅素Hb、紅血球比容Hct與白血球WBC)之影響。本實驗以三種濃度的丁香精油(200、300及400 ppm)分別在20℃與25℃下進行試驗。為了評估對波斯鱘使用麻醉劑後的生理反應,將波斯鱘移入含300 ppm丁香精油的25℃溶液中,出現麻醉效果(鰓蓋停止活動)後立即移入復甦水槽,分別於移入復甦水槽後的第0、1、6、24與72小時採血樣分析。另外,波斯鱘分別以20與25℃及三種濃度的丁香精油(200、300及400 ppm)進行麻醉處理,結果都在1.5分鐘之內出現鰓蓋不動的麻醉效果。波斯鱘經麻醉處理再移入復甦水槽72小時後,血中腎上腺皮質醇與葡萄糖濃度已經與對照組無顯著差異,血液參數的變動也恢復得與對照組無顯著差異。實驗結果顯示,使用丁香精油對波斯鱘進行麻醉的效果,以400 ppm及水溫24℃時最佳,麻醉後的復甦也較快。

In order to examine the efficacy of clove essence as an anesthetic on Persian sturgeon, Acipenser persicus, we conducted an experiment. We also investigate if clove essence anesthesia suppressed the normal plasma cortisol and glucose level increase in this species or not. The effects of clove essence on hematological factors (Hb, Hct, and WBC) was examined as well. Three different concentrations of clove essence (200, 300, 400 ppm) combined the two water temperature (20℃ and 25℃) were assessed. From one trial (concentration: 300 ppm and temperature: 25℃), blood samples were taken on 0 (immediately upon placement in recovery tank), 1, 6, 24 and 72 hours post recovery in order to evaluate physiological responses. Anesthetizing with different concentrations of clove essence (200, 300, 400 ppm) combined the water temperature (20℃ and 25℃) rapidly occurred in less than 1.5 minute after exposure to the clove essence. After 72 h both blood glucose and cortisol reach the level similar to the control group which was before applying clove essence. Hematological parameters after some fluctuation reached its level the same as control group of the experiments. With regard to anesthetic effects of clove essence dosages, the combination of 400 ppm and 24℃ was the best treatment for anesthetizing as well as recovery.
