Obesity in Rats without Hyperphagia Following Hypothalamic Operations

大白鼠在下視丘破壞及食量限制情況下之肥胖現象

摘要


訓練大白鼠每天僅進時三小時。俟其習慣後,記錄各鼠每天三小時所耗食量。二週後以外科手術破壞下視丘腹內側核,留一部未接受手術者作對照。此後,此九十八隻鼠分爲三組:第一組,42隻下視丘破壞鼠每天仍僅自由進食三小時,第二組,12隻下視丘破壞鼠,每鼠之飼料較開刀前平均多兩公分,每天仍飼以三小時;第三組,44隻正常對照鼠,其中28隻每天僅飼三小時,16隻每天飼3—6小時。手術後第三十天犧牲各鼠,測各鼠體脂肪含量及股骨長度則見:第一組中有十三隻鼠變胖,其三小時食量較正常對照鼠爲多;第二組中有六鼠變胖,其食量與正常對照鼠相等。正常鼠飼3-6小時其每天食量可與第一組胖鼠相等但却不變胖。此結果證明下視丘腹內側核破壞後之大白鼠可不呈貪食現象而變胖,故變胖可能並非貪食之結果。

Rats were trained to eat for only 3 hours daily. After training daily food intakes were measured for 2 weeks, and hypothalamic lesions were then placed. After operation, rats were divided into 3 groups. Group 1(42 lesioned rats)continued on the 3-hour feeding program. Group 2(12 lesioned rats)were given daily 2 grams more than they ate in the preoperative control period. Group 3(44 control rats, subdivided into 2 group)continued either on the 3-hour feeding program(28 rats)or were fed 3 to 6 hours daily so as to increase their daily food intake(16 rats).At the end of the 30th postoperative day all rats were sacrificed and their body fat content and femur length were measured. Linear growth was judged by nose-occipital length at the time of operation and at sacrifice. Feeding pattern was studied by measuring first hour food intake. Thirteen of the 42 lesioned rats of group 1 and 6 of the 12 lesioned rats of group 2 contained body fat high than 10% and were considered obese. Such obesity cannot be considered to be the result of hyperphagia, and is considered as a primary metabolic effect of the lesions.
