Effects of in Vivo GnRH Immunoneutralization on the Hypothalamic GnRH and Pituitary and serum LH in the Ovariectomized Rat

大白鼠於去卵巢後第40天,由腹腔注射0.5或2毫升自製之性釋素抗體。對照組僅注射2毫升之正常兔血清。注射後第4天由心臟採血。所有動物於注射後第15天犧牲。血清及腦下腺前葉的促黄體素以及下視丘之性釋素濃度以放射免疫法測定。注射後第4天,0.5毫升抗體處理組之血清促黄體素濃度此對照組低68%,但是2毫升抗體處理組之血清促黄體素濃度比對照組高231%。注射後第15天,三組間之血清促黄體素濃度無差異。0.5毫升抗體處理組之腦下腺促黄體素濃度未見改變,但下視丘的性釋素濃度升高65%。2毫升抗體處理組的腦下腺促黄體素濃度顯著地下降(40%),下視丘性釋素濃度亦此對照組高37%,但不顯著。這些結果顯示(1)藉抗體消解性釋素,可以使去卵巢鼠的促黄體素合成作用降低,(2)在去卵巢鼠中,促黃體素之合成與分泌似各自獨立進行。

A single ip injection of a rabbit antiserum to the synthetic gonadotropin-releasing hormone(GnRH)was given to the rats ovariectomized 40 days earlier. The blood samples were collected before and 4 days after the injection of anti-GnRH serum (Ab). Fifteen days after injection, all rats were sacrificed. The LH levels in the anterior pituitaries (APs) and sera as well as the GnRH concentrations in the hypothalami were measured by radioimmunoassay (RIAs). The serum LH was significantly lower in 0.5 ml Ab-treated ovariectomized rats than in normal rabbit serum(NRS)-treated animals on day 4 after injection. On the 15th day after passive immunization to GnRH a declined pituitary LH associated with an enhanced hypothalamic GnRH were observed in Ab-treated rats as compared with those in NRS-treated animals. However, no significant difference between the serum LH in Ab-treated and that in NRS-treated rats on the 15th day after injection. These results suggest that 1) the synthesis of LH in ovariectomized rats can be reduced by immunoneutralization of GnRH; 2) LH synthesis and release occur independently in ovariectomized rats.
