Continuous Hot Pressurized Fluids Extraction of Isoflavones and Soyasaponins from Defatted Soybean Flakes

本研究探討連續式熱壓流體萃取脫油黃豆粉中五種異黃酮及總皂苷。實驗結果包含萃取壓力(413-4410kPa),萃取溫度(333-393K),萃取流速(10-25mL/min),乙醇水比例(0-95%)與黃豆粉進料(80-450克),對五種異黃酮及總皂苷回收率的影響。並以田口實驗設計法,蒐尋萃取壓力,萃取流速,乙醇水比例與黃豆粉進料四變數影響回收率的最適萃取條件。實驗數據與歸納的結果指出,萃取流速與黃豆粉進料量均大大地影響五種異黃酮與總皂苷的回收率。進料越低,回收率越高。流速越高,回收率越高。當80%之熱壓乙醇在溫度383K,壓力551kPa與流速25mL/min條件時,可自80克的脫油黃豆粉中,回收95%的異黃酮與76%的皂苷。最後,以Amberlite XAD 16-HP樹脂管柱,分離純化異黃酮萃出液。五種異黃酮的純度,由萃出液乾燥物的1%,提高至純化物的58%,且五種異黃酮在純化程序的回收率達到99%。

This work examined the effect of pressure (413-4410kPa), temperature (333-393K), solvent flow rate (10-25mL/min), ethanol-to-water ratio (0-95%) and the feed loading (80-450g) on hot pressurized fluids extraction of isoflavones and soyasapoonins from defatted soybean flakes. A four-factor Taguchi experimental design was employed to optimize operational conditions of these extractions. Experimental results indicated that 95% of isoflavones and 76% soyasaponins were recovered from defatted soybean flakes by using hot pressurized 80% ethanol extraction at 383K, 551kPa and 25 mL/min with a 80 g feed loading. The recovered isoflavones were further purified by a solid-liquid column packed with Amberlite XAD 16-HP resin. The content of isoflavones in the collected fraction could be enhanced from 1 to 58% by weight.
