免疫酵素分析測定臺灣馬鈴薯病毒之發生與優良品系之抗Y病毒反應

Occurrence of Potato Viruses in Taiwan and Resistance to Virus Y of Some Promising Clones as Determined by ELISA

摘要

利用酵素結合抗體免疫分析(ELISA)檢測馬鈴薯病毒A(PVA)、S(PVS)、X(PVX)、及Y之嵌紋型系統(PVY-M)和壞疽型系統(PVY-N)在臺灣馬鈴薯產區之發生情形，結果顯示病毒發生並無地區性差異，而在於種薯的來源和品種（系）的不同。本所選育的優良品系No.143、No.770、385095-15、385095-54等皆普遍帶有PVS和（或）PVX，而較少PVY的發生，品系中No.143的PVY-M檢出率尤其偏低；但栽培品種卡地娜和克尼伯則少PVS或PVX的感染，而對PVY有較高的再感染率。在相鄰的試驗田，比較經檢查的當季種薯（豐農種）與自行留種的種薯所種出的克尼伯植株，前者生長勢強且病毒發生率比後者低，尤其是PVA和PVS。篩檢試驗田採收的薯球，因其中多數被PVY感染，在原來合格種薯長成的卡地娜樣本中已難尋得無病毒種薯存在；同時調查結果顯示PVY-M被檢出率比PVY-N高。在田間接種PVY-M或PVY-N進行抗病性檢定，結果顯示No.143品系對PVY（特別是PVY-M）的抗病性明顯大於卡地娜）。

Occurrence of potato viruses A (PVA), S (PVS), X (PVX), Y mosaic strain (PVY-M) and Y necrotic strain (PVY-N) in potato growing areas of Taiwan were investigated by ELISA. The frequency of virus detected from sampled locations showed no regional difference, but souces of seed-tubers and varieties cultivated made difference. All promising potato clones of No.143, No.770, 3865095-15, and 3865095-54 were found to be widely contaminated by PVS and/or PVX but much less infected by PVY. Particularly clone No. 143 was rarely infected by PVY-M. Cultivars Cardinal and Kennebec were frequently detected to be infected by PVY but seldom found to be infected by PVS or PVX. Comparing the potato plants from two neighboring fields, plants derived from current-year certified seeds outgrew those derived from second-year seed tubers, the latter being highly infected by viruses, especially PVA and PVS. Much more PVY-M than PVY-N was detected in tubers harvested from various experimental fields, indicating that mosaic strain is the dominant strain of PVY in Taiwan. High percentage of PVY reinfection to seed-certified potato cv. Cardinal in the field, resulted in very low rate of virus-free tubers produced. The results of inoculation with PVY-M or PVY-N to test the disease responses of clone No. 143 and cv. Cardinal indicated that clone No. 143 was more resistant to PVY infection (PVY-Min particular) than Cardinal in the field trial.
