以逢機增殖多型性DNA分子標誌鑑別番椒屬野生馴化種原

Study on the Molecular Markers of Capiscum Wild/Domesticated Species Using Random Amplified Polymorphic DNA Analysis

摘要


本試驗以源自日本、美國、巴西和匈牙利等地區之番椒屬14個品系,探討利用逢機增殖多型性DNA(RAPD)建立種間之分子標誌,以鑑別番椒屬重要野生馴化種之種原。試驗結果顯示,以UBC327, UBC346, UBC441及UBC457等四個隨意引子,經聚合酵素連鎖反應(PCR),其RAPD之產物具有明顯的多型性表現,且在增殖之多型性DNA片段中,可獲得11個有效之多型性DNA片段,提供作為不同「種」辨識之標誌。其中UBC327, UBC346及UBC457三個引子產生的多型性DNA片段,更具有指紋比對之功效,可以清楚地將14個不同品系加以區別。利用上述四個引子產生之所有多型性DNA片段,一併進行相似性群集分析(UPGMA cluster analysis)之結果顯示,供試的14個品系可分成四個群,第一群包含2個品系,屬於C. chinense;第二群包含4個品系,屬於C. annuum;第三群包含5個品系,也屬於C. annuum;第四群則包含3個品系,屬於C. baccatum。試驗結果證實,RAPD標誌對番椒屬具有指紋比對之效,可作為一種有效且值得信賴之種原鑑別工具。

Fourteen pepper (Capsicum spp.) accessions, originated from Japan, USA, Brazil, and Hungary etc. were surveyed by random amplified ploymorphic DNA (RAPD) analysis. The results indicated that high degree of polymorphism was obtained from four random decamer primers (UBC327, UBC346, UBC441, and UBC457) which produced 11 reproducible and effective amplification fragments useful for identification between species. Besides, each of the tested accessions of Capsicum could be exactly distinguished from each other by using the primers of UBC327, UBC346, and UBC457. Results of smilarity analysis cluster analysis (UPGMA, Unweighted Pair-Grouping Mean Arithmetic Averages) revealed these accessions could be divided into four groups: the first contained 2 accesssions, belonging to C. chinense, the second was composed of 4 accessions, belonging to C. annuum, the third contained 5 accessions, also belonging to C. annuum, and the fourth had 3 accessions, belonging to C. baccatum. The results strongly suggested that RAPD markers possessed the effect of fingerprintig and could be effectively and reliably used for classification Capsicum spp..
