1988年3月31日,臺灣新竹地區發生一件食魚中毒之事件,患者2名,其中1名死亡。患者之症狀爲舌頭、口唇、手脚乃至全身產生麻痺,並有呼吸困難之現象,復原者係經治療1週後痊癒,死亡者則一直陷於無知覺狀態,直到5月3日才死亡。這些症狀與河魨毒中毒者極爲類似。因此經依河魨毒之毒性生物檢定法測定魚體肌肉之毒性,得知爲每公克含有45個老鼠單位。爲鑑定食中毒之成分,乃將魚肉以1%醋酸甲醇溶液抽取後,經脫脂、超過濾和Bio-Gel P-2管柱層析法精製而得到粗毒,此粗毒經薄層分析法、醋酸化纖維膜之電泳分析法和逆相高性能液態層析儀等分析,得知含有河魨毒tetrodotoxin和其關連物質anhydrotetrodotoxin。殘留魚體之骨格,其椎骨形態相似於鯖河魨屬Lagocephalus,且其尾鰭形態呈新月形,因而鑑定其種別爲栗色河魨Lagocephaluslunaris。綜合上述,這次食中毒之毒成分係爲河魨毒所致,食中毒魚類則爲有毒種栗色河魨L.lunaris。
Attempts were made to elucidate the responsible toxin and identify thespecies in fish which recently caused food poisoning in Hsinchu County,Taiwan.The causative fish muscle was assayed for its toxicity as tetrodotoxin.Thescore of toxicity in fish muscle was 45 mouse units per gram.The toxin waspartially purified from the methanolic extracts of the fish muscle by ultrafiltrationand Bio-Gel P-2 column chromatography.Analyses of thin-layerchromatography,cellulose acetate membrane electrophoresis and high performanceliquid chromatography demonstrated that the toxin consisted of tetrodotoxinand anhydrotetrodotoxin.According to the morphology of the hemaland neural spines,the fish was identified to be a species of genus Lagocephalus(family Tetraodontidae).Furthermore,the fish possesed prominant lunatecaudal fin which is a unique character of the Lagocephalus lunaris.Hence,thefish was confirmed as L.lunaris.It was concluded that the causative agentof the above food poisoning was tetrodotoxin,and the fish species was atoxie puffer L.lunaris.
