Genetic Similarity of Geographic Caulerpa(Chlorophyta)Populations in Taiwan Island Revealed by Random Amplified Polymorphic DNA

摘要


1996年在臺灣中部的美洲鰻(Anguilla rostrata)養殖場爆發鰻魚鰾線蟲症,導致大量養殖鰻魚死亡。經解剖疑似感染鰻魚,觀察自鰾囊中游離出的成熟線蟲後發現,該線蟲具有強壯肌肉的食道與圍口齒的數目,因此鑑定該線蟲為粗厚鰻居線蟲(Anguillicola crassus)。在本實驗中,利用經常使用於寄生蟲蟲卵與幼蟲檢查的利用硫柳汞-碘-福馬林法(MIF)來檢查感染鰾線蟲的鰻魚。檢視後,MIF法的專一性與陽性預測率均為100%,但敏感度與陰性預測率略低,分別為56%與36%。綜合考慮後,MIF法具有方便便宜與易於推廣的特性,因此值得應用於一般的養殖場進行鰾線蟲的篩檢。

In 1996,anguillcolosis were observed causing a mass mortality of the intensive Americaneel,Anguilla rostrata,aquaculture facility in central Taiwan.Examining the matureadult worms from the lumen of swim bladder in infected eels,the nematode was identifiedas Anguillicola crassus based on the morphology of esophagus of adult worm and thenumber of circumoral teeth(n=25).Merthiolate-iodine-formalin(MIF)stain method,agood stain preservative for most kinds and stages of parasites found in feces,is used toobserved the eels infected by Anguillicola in this examination.The specificity and predictivevalue positive of MIF were both 100%,and the sensitivity and predictive value negativewere 56% and 36% respectively.The method of MIF is cheap and convenient to bepromoted in examining of anguillicolosis in aquaculture facility.
