培養基成分對甘藷組織培養生産花青素之影響

摘要


本研究以因子試驗探討2,4-D及kinetin對誘發甘藷紫紅色癒合組織之影響,於試驗範圍內kinetin對其紫紅色程度有極顯著影響,2,4-D則無;kinetin濃度爲0.1、0.5、1.0或2.5μM時,其癒合組織紫紅色程度較高。 利用三因子三層階之1/3部分複因子試驗探討培養基中蔗糖、氮源及磷酸鹽濃度對癒合組織紫紅色程度之影響,試驗結果以蔗糖有極顯著影響,而氮源及磷酸鹽則無。以中心混成之反應曲面法探討蔗糖與kinetin對癒合組織紫紅色程度之影響,雖並未尋行最適點,但kinetin濃度在0.6~3.4μM間,濃度愈低可産生紫紅色程度較高之癒合組織;蔗糖則以2.3~3.2%(w/v)之紫紅色程度最高。 以五因子五層階之1/4部分複因子試驗探討培養基中蔗糖、氮源、磷酸鹽、2,4-D及kinetin對甘藷懸浮培養細胞生長之影響:氮源、磷酸鹽及2,4-D有極顯著影響;蔗糖及kinetin則無。以中心混成之反應曲面法探討氮源、磷酸鹽及2,4-D對懸浮培養細胞生長之影響,結果並未尋得最適點,但在試驗範圍內2,4-D及氮源的濃度愈高,細胞生長愈多。

Factorial experiment was used to study the effects of 2,4-D and kinetin on the initiation of purple call from the leaf explants of sweet potatoes, Once the ranges tested, the kinetin significantly affected the degree of purple of calli initiated while the 2,4-D did not. Using a 1/3 replication of a three-factor, two-level fractional factorial experiment to investigate the effect of concentration of sucrose, nitrogen and phosphate on the degree of purple of colts initiated. Results showed that the concentration of sucrose significantly affected the degree of purple of calli while these of nitrogen and phosphate did not. The response surface methodology with a central composite design was used to measure the effects of the concentration of sucrose and kinetin on the degree of purple of cults. Although the optimum concentration of sucrose and kinetin was net found, 0.6 to 3.4 μM of kinetin or 2.3 to 3.2% of sucrose did enhance the degree of purple of calli. Using a 1/4 replication of a five-factor, two-level fractional factorial experiment to study the effects of sucrose, nitrogen, phosphate, 2,4-D and kinetin on the growth of the cells in the suspension culture, it was found that the concentration of nitrogen, phosphate and 2,4-D did significantly affect the growth. However, the concentration of sucrose and kinetin did not show n significant effect, The response surface methodology with a central composite design was used to measure the effects of the concentration of nitrogen, phosphate and 2,4-D on the growth of suspension cells. The optimum concentration of those factors were not found, hot over the experiment ranges the higer the concentration of 2,4-D and nitrogen the mare the growth of cells.
