本研究目的為評估柳杉(Cryptomeria japonica D. Don)葉部精油的抗細菌活性,同時由精油中分離與鑑定具抗細菌活性的成分。試驗結果顯示柳杉葉部精油對於金黃色葡萄球菌(Staphylococcus aureus)與糞腸球菌(Enterococcus faecalis)的最低抑制濃度分別為2mg/mL與1mg/mL。進一步由活性分離部中分離得到ent-Kaur-16-ene、γ-Eudesmol、β-Eudesmol、β-Elemol、α-Cadinol與cis-Sabinene hydrate,其中,cis-Sabinene hydrate為首次自柳杉中分離與鑑定得到的化合物。由精油成分的抗細菌活性評估,得知β-Elemol、α-Cadinol與ent-Kaur-16-ene的抗細菌活性較佳,對於抗藥性金黃色葡萄球菌(Methicillin-resistant Staphylococcus aureus, MRSA)的最低抑制濃度均未超過0.5mg/mL。綜合上述結果可知柳杉葉部精油具有良好的抗細菌活性。
The objective of this paper was to evaluate the antibacterial activity of leaf essential oil from Cryptomeria japonica D. Don. Minimum inhibitory concentration (MIC) values of leaf oil were 2mg/mL against Staphylococcus aureus and 1mg/mL against Enterococcus faecalis respectively. Six terpenoids including cis-sabinene hydrate, γ-eudesmol, β-eudesmol, β-elemol, α-cadinol and ent-kaur-16-ene were isolated and identified from active fractions of leaf oil. Among these compounds, cis-sabinene hydrate was isolated for the first time from this plant. Results from antibacterial assay revealed that β-elemol, α-cadinol and ent-kaur-16-ene exhibited inhibitory effects against bacteria. MIC values of these three constituents were all less than 0.5mg/mL against methicillin-resistant Staphylococcus aureus (MRSA). Results obtained demonstrated that leaf oil of C. japonica possesses the commendable antibacterial activity.
