以ITS分子序列診斷樹木靈芝病害

RDNA-ITS SEQUENCES AS AN AID TO DIAGNOSIS GANODERMA DISEASE OF TREES

摘要


靈芝是重要的藥用真菌,但也是台灣目前僅次於樹木褐根病菌外,造成樹木病害的重要病原菌。靈芝的鑑定主要靠子實體形態上的特徵,但若被感染的樹木已經開始出現病徵,卻未見子實體的形成或是子實體形成後遭人移除,這些都會造成確診的障礙。有經驗的專家可透過採取病健部進行培養鑑定,根據菌落進行較準確的推測,但是其準確性僅可以到達屬,卻無法僅憑培養菌落鑑定到達種;透過太空包的子實體形成法是可以做出正確的診斷方法,惟需費時數月的培養時間。因此本研究嘗試以選擇性培養基培養法進行病原菌的分離純培養,然後再以ITS分子序列進行解序比對分析,可以在未發現子實體的情況下獲得快速而正確的診斷結果,提供日後相關人員鑑定上的參考。

Ganoderma spp. have extensive use in traditional Chinese medicines, however, following the top tree killer Phellinus noxius, they are the second most important tree pathogen group in Taiwan. The Diagnosis of Ganoderma diseases mostly rely on the identification of fruiting bodies, which may not be present in some cases. Only few experienced researchers can identify them by the morphology and microscopic structure of mycelium, and only at the genus level, but not at the species level. Fruiting bodies can be cultivated from the isolated mycelium, but it may require long time for incubation. Aiming at fast and accurate diagnosis of Ganoderma diseases, this research studies the potential of rDNA analysis with ITS primers on the mycelium of pathogen isolated from the infected trees with selective media.
